| Ophiopogon japonicus(Thunb.)Ker-Gawl is a perennial herb of Liliaceae.It is a famous traditional Chinese medicine.Paclobutrazol is very popular with farmers,because it has the function of inhibiting the apical dominance,promoting the root expansion and increasing the yield of Ophiopogon japonicus.In recent years,with the spraying of Paclobutrazol and the properties of Ophiopogon japonicus rotation,the residue of Paclobutrazol in Radix Ophiopogon is far beyond the national standard.In this study,the effects of different fertilization treatments on the biological characters,quality and and the residue of Paclobutrazol were compared in order to provide scientific basis for the planting and quality standard of green Ophiopogon japonicus.The main results are as follows:1.The biological characteristics of Ophiopogon japonicus by different fertilization treatments.were studied,the results showed that the biological bacteria fertilizer had a certain improvement on the overall quality of Ophiopogon japonicus.The effects of light carbon nuclear fertilizer on the biological characters and yield of Ophiopogon were mainly in the upper part of the ground.The growth rate of the biological fertilizer to the yield and the high quality root growth rate of Ophiopogon japonicus was stronger than that of Paclobutrazol and light carbon nuclear fertilizer(30.45% > 14.87% > 1.48% and 31.43% > 21.2% >-29.66%).2.A preliminary TLC study of Ophiopogon japonicus under different fertilization treatments was carried out.The TLC results showed that the samples of Ophiopogon japonicus have the same spots in the same position on laminate,indicating that the two components were contained in the samples of Ophiopogon japonicus.According to the requirements of People’s Republic of China Pharmacopoeia(2015 Edition),Ophiopogon japonicus requirement.3.The effective components of Ophiopogon japonicus by different fertilizer treatments were determined.The results showed that the application of bio bacterial fertilizer increased the content of aqueous extract alcohol extract,total polysaccharide,total flavonoids and total saponins in Ophiopogon japonicus,which increased by 2.11%,7.33%,7.06%,22.83% and 6.57%,respectively.The effect of light carbon Hefei on the accumulation of root leach,total polysaccharide and total flavonoids in Ophiopogon japonicus increased by 1.31%,1.62%,9.86% and 8.62% respectively,and the effect on saponins could not be determined for the time being.Ecological fertilizer enhancing the content of aqueous extrac and total flavonoids as well as total saponins of Ophiopogon japonicus higher than Paclobutrazol and light carbon fertilizer.On Pharmacopoeia index,application of bio fertilizer on quality to enhance the quality have the best effect of Ophiopogon japonicus.4.The results showed that the increase between the flavonoids and saponins in root of Ophiopogon japonicus was higher than that of fibrous root and blade(0.87% > 0.71% > 0.18% and 0.74% > 0.56% > 0.18%).The increase among the extracts is that the tuberous root is higher than the fibrous root and higher than the leaf(alcohol extract: 80.41% > 52.21% > 35.91%,alcohol extract: 11.46% > 8.51% > 7.43%).The total polysaccharide of fibrous roots was higher than that of root tuber and higher than that of leaves This study can provide a scientific basis for the development and utilization of fibrous root and leaves of Ophiopogon japonicus.5.Detection of Paclobutrazol Residue in Ophiopogon japonicus by different fertilization treatments and its soil was carried out.The results showed that there was a significant positive correlation between paclobutrazol residues in Ophiopogon japonicus and soil with the application of Paclobutrazol(P<0.001***).The residual amount of Paclobutrazol in different parts of Ophiopogon japonicus was significantly different,and the residual amount of Paclobutrazol in fibrous roots was about 3~4 times higher than that of root and fibrous root.The residue of Paclobutrazol in soil was about 7~10 times higher than that of high quality root tuber and fibrous root.Application of bio fertilizer can significantly optimize soil micro ecological environment,and it has a certain degradation effect on the Paclobutrazol of Ophiopogon japonicus plantlets and soil,the average rate of degradation is 91.52%,and its degradation mechanism needs further exploration.6.Microbial community tests were carried out on soil with application of Paclobutrazol and blank control soil.It was found that Paclobutrazol promoted the reproduction of bacteria and fungi in the soil,and the multiplication of multiple actinomycetes was inhibited.Though Paclobutrazol played a significant role in enhancing the biological characteristics,yield and effective components of Ophiopogon japonicus,the ecological environment pollution caused by Paclobutrazol should not be ignored.By comparing and analyzing the effects of different fertilization on the quality of the sensory quality and quality of Ophiopogon japonicus,and consideration of ecological environment and food safety,proposing to joint applied bio fertilizer and light carbon fertilizer in the process of planting green Ophiopogon japonicus.Through the experimental research and analysis and evaluation of this topic,the quality standard of green Ophiopogon japonicus was drafted,and the technical regulations for the production of green Ophiopogon were drawn up,which provided an important scientific basis for the comprehensive
