The triangle maple is a deciduous tree of Acer buergerianum Miq,which is a good native maples in our country.Trident maple strong adaptability,resistance to drought or poor,and is also resistant to certain water wet,the trident maple wood is good,has the certain medical value,can also be used as mixed corrosion inhibitor,and trident maple is important color leaf tree species and forest scenic resources.At present,there is less research on the triangular maple in China,mainly focusing on the application of gardens,especially in the field of breeding.Polyploid is an important parent material for breeding new cultivars,and it is of great significance to the discovery of the polyploid of the triangle in germplasm resources.With the selection of new clones and varieties,it is necessary to study the DNA fingerprinting of the triangular maple.This test by breeding new four triangular maple clones and 2 sands maple clones for research object,using flow cytometry instrument identified six clone times,and is verified from the aspects of morphology,and build the DNA fingerprint,for parent selection,variety identification,breeding and so on has laid a solid foundation.The results of this experiment are as follows:1.The ploidy test of plants was carried out by flow cytometry.In this experiment,we compared the effect of 3 kinds of lysate on the nucleus suspension of the seedlings of Acer tran.The result showed that the 1 lysate had the best effect,less impurities and higher resolution.The preparation process of nuclear suspension was also optimized,and the final detection result was good.Compared with the diploid triangulation maple,6 clones ploidy was identified by flow cytometry.The peak of the trigonometric Maple No.1 clones was 2times of the diploid,and the other 5 clones were the same as the diploid.The results of flow cytometry showed that the single cell DNA content in the leaves of No.1 clones was 2 times more than that of diploid,and the DNA content of the other 5 clones and diploid seedlings was similar to that of the single cells of the diploid seedlings.2.The electron microscopic observation showed that the stomatal length of the triangle Maple No.1 clones was 38.51% larger than that of No.2 clones,and the stomatal density was33.57% smaller than that of No.2 clones,and the paraffin section showed that the palisade tissue,the lower epidermis cells and the guard cells were larger than the 2 clones in the triangle Maple 1,and the morphological aspect,the triangle Maple 1 clone was compared with the 2 clone.There were significant differences in leaf width,leaf length,leaf thickness,petiole length and petiole width.There are obvious differences in stomata,leaf shape,leaf anatomy and DNA content in the 1 and 2 clones of maple maple,which can be identified as tetraploid.This is the first natural polyploid variation of trigonometric maple.It is of great significance to the study of the germplasm evolution and breeding of the triangle maple.The cold hardiness of different strains was different.The cold resistance of 6 clones was determined by conductance method.The order of strength was J1>J2>S1>S4 S2>S3.The cold resistance of J1 and J2 is obviously higher than that of other clones.This is also consistent with the characteristics of the 6 clones that are observed.It provides the material for the cultivation of the cold resistant evergreen plantation in the north,and is also of positive significance against the cold introduction.3.There are extensive genetic polymorphisms among the clones of Acer buergerianum Miq.The fingerprints constructed by SRAP markers can distinguish the genotypes of the 6clones,which are easy to operate and have strong stability,and can be used to identify varieties and strains quickly.The 3 pairs of primer combinations(Me1/Em4,Me7/Em8,Me8/Em9)selected in this experiment can accurately identify S1,S2,S3,S4,J1 and J2 in clones,and the confidence rate is higher than 99.99%.The DNA-SRAP fingerprint was established for the single plant of fine Acer buergerianum Miq triangulation,which provides some reference for molecular identification,germplasm identification,genetic resources management and variety protection of fine single plant of maple Acerpaxii Franch and Acer buergerianum Miq.
