Panax notoginseng belongs to the genus Panax,Araliaceae,which is an important traditional Chinese medicine in China.It mainly grows in Wenshan,Yunnan Province,China,with a long history of cultivation.Previous studies showed that P.notoginseng has the effects of hemostasis,analgesia,lowering blood pressure,regulating blood glucose,improving immunity,and so on.To meet the growing market demand it is needed to explore germplasm resources of P.notoginseng through tissue culture and other breeding technologies to improve the quality of this medicinal plant.In this study,we attempted to obtain methods for the tissue culture and polyploidy induction in P.notoginseng.The major results are as follows:1.For the callus induction,the optimum explants for callus was stems.We found that steMS serves as the best disinfection method with 75%alcohol disinfection for 30 seconds and 5%sodium hypochlorite disinfection for 30 minutes.The best browning inhibitor and concentration of PVP 0.05g/L,the best callus induction medium was MS medium + 3.5mg/L 2,4-D+ 0.8 mg/L KT.2.For the callus differentiation induction,the optimum culture medium of callus adventitious root differentiation was MS + 1 mg/L 2,4-D,while the optimum medium of adventitious bud differentiation was MS + 1.5 mg/L GA3+ 1.5 mg/L 6-BA.3.We failed to find a proper method for the polyploid induction of callus,but our experimental results showed that that colchicine can affect callus growth,that is,low concentration of colchicine may have effects on callus chromosome doubling.Trident maple Acer buergerianum Miq.,a member of the family of Aceraceae.As an important ornamental tree it has enormous commerce values.In recent decades human activities have seriously destroyed natural habitats,making the loss and quick decline of a large number of natural populations of wild trident maple in China.We report the completed chloroplast genome sequence of Acer buergerianum using next-generation sequencing technology.The complete chloroplast genome size is 156,477 bp.The genome contained 134 genes,including 89 protein-coding genes,40 tRNA genes and 8 rRNA genes.The most of genes occur as a single copy,while 23 genes occur in double copies.A phylogenetic analysis based on chloroplast genomes indicates that A.buergerianum is closely related to A.miaotaiense,A.morrisonense and A.davidii.The complete chloroplast genome of A.buergerianum build a good foundation for bio-diversity conservation,molecular and ecological studies.
