At first,this paper took the Early-maturity Jing Qie NO.1,Jing Qie NO.218,Jing Qie Hei Longwang,Hei Ba,San Ya NO.5 and 1112 x 407 six different varieties of eggplant seeds as the experimental materials.Researched on the different concentrations of the tetrazolium solution,staining temperature and staining time for the determination of eggplant seeds viability,figuring out the optimum conditions of TTC staining method for determining eggplant seeds viability.Aims to provide references for the eggplant seeds production and sales,for the determination of eggplant seeds viability by TTC staining method as soon as possible included in China's “Crop Seed Testing Rules” and the“International Rules for Seed Testing” lay the foundation.Secondly,to explore the influencing factors of breaking seeds dormancy in the germination.This paper specially chose Solanum Torvum seeds as the research material,which produced by CHENGDU HAOTE YUANYI YOUXIAN GONGSI in 2014,without breaking dormancy processing has strong characteristics of dormancy.Carried out the analyzed on the basis of existing research,a variety of methods have been used to try to break the seeds dormancy,in order to obtain the suitable method for breaking Solanum Torvum seeds dormancy and apply to agricultural production.Meanwhile,in order to further explore during the seeds dormancy release the changes of enzyme activity related to cell wall degradation.In the process of Solanum Torvum seeds dormancy release,?-mannanase,?-mannosidase and ?-galactosidase activity changes were studied.The results as follows:1.Soaking seeds in water at 20? for 18 h,concentration of the tetrazolium solution was1%,staining at 35? for 18 h,which was the suitable TTC staining method for rapid determination of eggplant seeds viability.2.Solanum Torvum seed has a strong dormancy characteristic,and the fresh harvested seeds were not germinated.3.30? was an ideal temperature of constant temperature germination,Solanum Torvum seeds began to germinate in the first 11 d,and 30 d the germination rate of 94%;Compared with constant temperature germination,changing the germination temperature was conducive to breaking Solanum Torvum seeds dormancy,20?16h/35?8h cycle process was most satisfactory,Solanum Torvum seeds began to germinate in the first 7d,and 14 d the germination rate of 96.25%.On the basis of appropriate temperature conditions to give suitable photoperiod,was more advantageous to breaking Solanum Torvum seeds dormancy,35? for 14 h with light(light source: incandescent light bulb;light intensity:750-1250 lux,the same below)and 20? for 10 h dark cycle conditions,better effect,Solanum Torvum seeds began to germinate in the first 6d,and 14 d the germination rate of95.5%,indicted that seeds dormancy has been released.4.Suitable concentration of GA treatment was beneficial to breaking Solanum Torvum seeds dormancy.With the concentration of 5.0m M or 10.0m M GA soaking seeds for 24 h can effectively breaking Solanum Torvum seeds dormancy,seeds began to germinate in the first 3d,and 14 d the germination rate reached 97.34% and 98%,respective.But the GA concentration was difficult to master,it's too low or too high with no effect and the high concentration easy to cause damage to the seeds,also extensive use of GA high cost,so suggested that large-scale production of Solanum Torvum seeds using 5.0m M GA solution to breaking the dormancy is more economical.The experimental results of some other plant hormones were not ideal,and to break Solanum Torvum seeds dormancy without too much significance.5.Low temperature stratification(4?)and room temperature ripening on Solanum Torvum seeds dormancy release of little effect.6.Solanum Torvum seeds were treated with common chemical reagents,without ideal effect to break the dormancy.7.In the process of Solanum Torvum seeds dormancy release,?-mannanase,?-mannosidase and ?-galactosidase activity changes were studied.In the dry seeds,?-mannanase activity was not detected,and ?-mannosidase and ?-galactosidase activity already exists;With the deepening process of germination,?-mannanase activity gradually increased,and before the seeds germinate soon its activity must reach a certain intensity,then the seeds began to germinate properly;?-mannosidase activity gradually increased;?-galactosidase activity first declined,then increased gradually,and before seeds germination upcoming the activity at a low level.
