Chinese wolfberry is the dry and ripe fruit of Lycium barbartum L.,which is one of the most valuable Chinese medicinal herbs in China.It is one of the first batch of medicinal and medicinal varieties in China.It has been used for more than 2000 years,Medicinal history,in recent years has become renowned at home and abroad tonic diet treasures.Lycium barbartum L.is tasted sweet,nature,nourishing liver and kidney,the purpose of clever purpose and regulate the body immunity,anti-aging,anti-fatty liver and other functions,by the version of "Chinese Pharmacopoeia" was included.Modern medicine,nutrition and pharmacology research found that wolfberry contains the human body needs polysaccharides,amino acids,vitamins,trace elements,are widely used in anti-aging,anti-tumor,and anti-cardiovascular disease Lycium barbartum L.has a very high medicinal value and nutritional value.In this study,I selected the polysaccharide and flavonoids from Lycium barbarum L.as raw materials,and provided the theoretical basis for its development and utilization.1.The content of Lycium barbarum polysaccharides prepared by high speed shearing was identified by color reaction and visible light spectroscopy.The method of high-speed shear Dispersing Emulsifier?HSDE?was used to extract the lycium barbarum polysaccharide [18-19].The effects of shear time,shear rate,ratio of material to liquid and extraction time were studied by single factor and orthogonal test,on the Extraction Rate of Lycium Barbarum Polysaccharides.The optimum extraction conditions were as follows: the extraction temperature was 60?,the shear time was 2 min,the shear rate was 12000 rpm,the ratio of material to liquid was 1:12,and the extraction time was for 2 h.Further optimize the alcohol precipitation process,the best conditions are: The ratio of raw material and extract is 1: 2,alcohol concentration 60%,alcohol precipitation time 2 h.Under the optimized extraction conditions,the extraction rate and product purity of Lycium barbarum were 36.19% and 48.61%,respectively.2.Purification of Flavonoids from Lycium Barbarum by Macroporous Adsorption.According to the literature reported rutin,quercetin,apigenin,luteolin and kaempferol is antioxidant,improve immunity and anti-tumor common Chinese wolfberry flavonoids.In this study,the content of flavonoids and the flavonoids of Lycium barbarum were determined by high performance liquid chromatography?HPLC?,and the molecular size and molecular polarity of these flavonoids were calculated.A series of macroporous resins?BSKB-1,BSKC-1,AUKJ-1,BWKS-1,etc?,and two kinds of macroporous resins?BWKX-1,AUKJ-1?were prepared by static adsorption and desorption.The mixing ratio of the two resins is preferably obtained by static adsorption and desorption.In addition,to determine its specific dynamic adsorption,desorption conditions are as follows: The concentration of flavonoids in wolfberry was 600 g·L-1,the flow rate was 10 BV·h-1,the selected eluent was 60% ethanol and the elution flow rate was 15 BV·h-1.Under the optimized conditions,the content of rutin was increased from 0.06% to 0.21%,and the content of myricetin increased from 0.08% to 0.23%.In this experiment,the separation and purification of flavonoids from Lycium barbarum L.was optimized,and the flavonoids of Lycium barbarum L.were isolated and the purification efficiency was improved.3.The optimal conditions of selenium,selenium were screened by single factor and best value screening and response surface analysis using atomic fluorescence and in vitro chemical reaction system.According to the addition of LBP as 1 g,adding 0.62 g Na2SeO3,with 1% HNO3 fully dissolved,then adding BaCl2 1 g,with oil bath 74? reaction 9 h.After completion of the reaction,the saturated Na2CO3 solution to adjust the pH to 5-6,Na2SO3 solution to precipitate Ba2+,centrifugal supernatant,with 1 kDa dialysis bag tap water for 2 d,distilled water for 1 d,concentrated and dried.Under the optimized extraction conditions,the selenium content of polysaccharide from Lycium barbarum was 648.65 ug·g-1.On the basis of this,the MTT method was used to evaluate the inhibitory effect of Lycium barbarum polysaccharide on HePG2 human hepatocellular carcinoma cells.In summary,this paper optimized the high-speed shear-assisted LBP extraction process and its alcohol precipitation conditions.The flavonoids of Lycium barbarum were isolated and purified by macroporous adsorption resin and the contents of rutin and myricetin were determined by high performance liquid chromatography?HPLC?.The content of rutin was increased by 2.5 times and 1.9 times;The optimum conditions of saponification of Lycium barbarum polysaccharides were optimized and the antitumor activity of Lycium barbarum polysaccharide was studied in vitro.For the further development and utilization of Chinese wolfberry functional food to provide a certain experimental basis and theoretical basis.
