The main topic of study is pay attention to the protection mechanism of rice residue protein peptides for human umbilical vein endothelial cells.Verified its antioxidant mecha-nism through three levels of cells,proteins,molecules.Make rice production process waste slag as raw material,based on preliminary findings of the Task Force on the screening of a short chain of rice active peptide,synthesizer by amino acids obtained in vitro experiments for high-purity peptides.Constructed in vitro injury model.Using MTT assay,cell flow cyt-ometry select the best protection time and concentration of rice peptides,and make the gro-wth curve,intuitive to determine the viability of apoptotic cells and other phenomena,and then assisted light microscopy,fluorescence microscopy Morphological observation using HE staining,Hoechst staining and morphological observation under growth conditions of intracellular organelles;DNA damage through comet assay,cell migration experiments angiogenesis qPCR experiments verify regulatory RNA signaling pathway,and finally Western blot analysis of vascular endothelial factor-related apoptosis-regulating mechanism of NF-?B in the case of injury.Experimental results shows that the impact of rice active peptide?RBP?injury induce-d by H2O2 in human umbilical vein endothelial cells,viability of the screened RBP at 0.3 mg/mL for 24 h is the best protective effect on HUVEC.Compared to natural Vc,its antioxidant activity of antioxidants or less;Morphological changes in cell structure shows RBP protection group and the normal control group similar to the intact cell structure,cell shape and stretch the rules of good,it has rich organelles,and its cell surface was smooth;no morphological injury group Rules nuclear the membrane was retracted,nuclear transfer spills,irregular organelles,cell damage in the state;apoptosis experiments showed that compared to the oxidative damage,the rice peptides protection group protective role in the group of more than 50%.Hollow cell migration assay showed that rice peptides protection group can form the shape of the rules of micro control,and oxidative injury group can not.qPCR,Western blot experiments showed that rice peptides reduced NF-?B protein express-ion by inhibiting apoptosis of HUVEC protection.The results showed that rice peptides can effectively protect HUVEC cell from oxidative damage,at the same time it also can improve the viability of HUVEC cells involved in apoptosis and downregulated factor NF-?B protein.Average water molecules from the cell to exhibit good oxidation activity.By the latter part of this research will be to develop depth of rice peptides provide strong theoretical support use.
