Pomegranate peel polyphenols are all the sum of polyphenol hydroxyl compounds in pomegranate peel,It contains gallic tannins,ellagic tannins,ellagic acid,chlorogenic acid,gallic acid,catechin,epicatechin,anthocyanin,ferulic acid and quercetin etc. It is about 10%-20% of it's dry weight.Many Studies show that pomegranate peel polyphenols have fight free radicals, induced resistance, anti-aging and bacteriostatic effect, it has a huge role in many fields such as medicine, daily necessities, food and so on.We had studied on the qualitative and quantitative analysis method of Pomegranate peel polyphenols,The extraction of pomegranate peel polyphenols?Pomegranate peel extract on ?-amylase, ?- glycosidase enzymes and inhibition of fatty acid synthase in vitro experiment,and we also studied the inhibition effect of pomegranate peel extract on the proliferation and differentiation of fat cells,we aimed to provide reference for the development of new non-side effects of weight loss drugs and different functional foods.The paper was studied from the following three parts.1 microwave assisted extraction of Pomegranate Peel Polyphenols extractObjective Extraction of polyphenols from pomegranate peel by microwave assisted extraction. Methods By means of single factor test and response surface optimization to get the best optimum technological condition for extraction of polyphenols from pomegranate peel. Results The best optimum technologic condition:Sieve mesh 120 mesh, feed liquid ratio 1:20, extraction time 78 s, microwave power low heat(264w), extraction series was 3, the yield of polyphenols crude extract was 23.26g/100 g.The pomegranate peel extracts were concentrated and freeze-dried, finally get the pomegranate jelly powder through the polyphenol content of calculation formula of the extraction rate reached 56.10%. Conclusion The method is more effective than most of the traditional methods, the operation is simple, and the extraction efficiency is high.2 The inhibitory effect of Pomegranate peel polyphenols extract, Punicalagin, ellagic acid on the ?- amylase, ?- glycosidase enzymes and fatty acid synthaseObjective To research the inhibitory effect of Pomegranate peel polyphenols extract, Punicalagin, ellagic acid on the ?- amylase, ?- glycosidase enzymes and fatty acid synthase. Methods By comparing the difference of the absorbance value between the sample group and the control group in the reaction system, to explore the effective concentration range of the sample to the inhibition of these enzymes, and calculate the semi inhibitory concentration. Results Pomegranate peel extract has obvious inhibitory effect on alpha amylase and alpha glucosidase, fatty acid synthase, the semi inhibitory concentration IC50 were: 0.7972mg/ml?0.2605mg/ml?74.13mg/ml, the semi inhibitory concentration IC50 of the ellagic acid on the alpha amylase and alpha glucosidase, fatty acid synthase were:0.0810mg/ml?0.0262mg/ml?0.55mg/ml, the semi inhibitory concentration IC50 of the Punicalagin on the alpha amylase and alpha glucosidase, fatty acid synthase were:0.0678mg/ml?0.0320mg/ml?0.054mg/ml. Conclusion According to the results of the semi inhibitory concentration,it can be seen that the pomegranate peel polyphenolic extract,ellagic acid Punicalagin have better inhibitory effect on alpha amylase, alpha glucosidase and fatty acid synthase in proper concentration,that is Pomegranate Peel Polyphenols can reach lipid-lowering by inhibiting the enzyme related obesity.3 Inhibitory effect of pomegranate peel extract and ellagic acid on the proliferation and differentiation of preadipocytesObjective To research the inhibitory effect of pomegranate peel extract and ellagic acid on the proliferation and differentiation of preadipocytes. Methods To observe the effect of pomegranate peel extract, ellagic acid on the proliferation of human preadipocytes by MTT method,the effect of pomegranate peel extract, ellagic acid on the differentiation of human preadipocytes by oil red O staining.Results In the inhibition ofpreadipocytes proliferation experiments,25?g/ml of pomegranate husk extract and 20?M of ellagic acid inhibitory effect o-n the preadipocytes is the best,the inhibition rate were 31.87%, 65.83%;In the inhibition of preadipocytes differentiation experiments,25?g/ml of pomegranate husk e-xtract and 10?M of ellagic acid inhibitory effect on the preadipocytes is the best,the inhibition rate were 45.88%?44.96%.Conclusion In a certain concentration range,pomegranate peel extract and ellagic acid has inhibitory effect on preadipocytes,Pomegranate peel polyphenols can inhibit the proliferation and differentiation of preadipocytes, so as to achieve the goal of reducing fat.
