The living standards of human beings are constantly changing,and the high-fat diet leads to an increasing incidence of chronic metabolic diseases,mainly non-alcoholic fatty liver disease(NAFLD),and the incidence rate shows an obvious younger trend.However,there is no ideal drug to treat it.Therefore,early intervention and reversal of the inflammatory response of the liver are critical to its treatment and prevention.In recent years,pomegranate peel polyphenols have been widely concerned by researchers for their nutritional and medicinal values such as antioxidant,anti-cancer,anti-inflammatory,anti-bacterial,lipid-lowering etc.In this study,a rat model of NAFLD induced by high-fat diet was established to investigate the alleviating effect of pomegranate peel polyphenols on NAFLD and its possible molecular mechanism.In this study,pomegranate peel polyphenols were identified by high performance liquid chromatography(HPLC)and the NAFLD model of SD rats fed with high-fat diet was established.The model group and the related drug group were fed a high-fat diet,and the animals in the drug group were fed pomegranate peel polyphenols(150 and 300 mg/kg)and simvastatin(1 mg/kg)daily for 8 weeks.The normal group and the pomegranate peel polyphenol high dose(300 mg/kg)control group were normal drinking water and intake.After 8 weeks,compared with the normal group,the weight of rats in the high-fat model group was significantly increased,and the serum and liver total cholesterol(TC)and triglyceride(TG)levels were significantly increased.Combined with HE staining analysis,when inflammatory cell infiltration occurred in the liver of SD rats in the high-fat model group,it indicated that inflammation occurred in the liver of rats.Combined with these experimental results,the modeling of NAFLD in SD rats fed with high-fat diet was successful.The levels/expressions of serum and liver inflammatory factors mRNA IL-1? and IL-18 and the activity/level of MDA,GSH-pX,SOD,T-AOC and CAT in rat liver were further detected;The relative expressions of TXNIP,NLRP3,SREBP-lc and FAS mRNA and protein in liver were determined.The research results are as follows:1.After 8 weeks of high-fat diet,the body weight of rats in the high-fat model group was significantly increased,and the serum and liver cholesterol(TC)and triglyceride(TG)contents were significantly increased.The results of HE staining indicated that inflammatory cell infiltration occurred in the liver of SD rats in the high-fat model group,which means the NAFLD model of SD rats were successful establishment in this study.2.Pomegranate peel polyphenols have antioxidant effects,which could effectively reduce the level of MDA in the liver and enhance the activity/level of GSH-pX,SOD,T-AOC and CAT.3.Pomegranate peel polyphenols can significantly reduce the production of IL-1?and IL-18 inflammatory cytokines and mRNA expression in the blood and liver of rats,and may slow down the development of NAFLD.4.Pomegranate peel polyphenols could reduce the expression of TXNIP mRNA and protein in the liver of rats,thereby inhibiting the activation of NLRP3 inflammatory bodies and thereby improving the inflammatory injury and lipid accumulation in the liver.5.Pomegranate peel polyphenols could reduce the expression of SREBP-lc and FAS mRNA and protein in the lipid metabolism pathway,and improved the lipid accumulation in the liver of rats,thus slowing down the development of NAFLD.In summary,this study showed that pomegranate peel polyphenols could reduce the NAFLD model rats induced by high-fat feed liver oxidative stress and inflammation,and through the lower the TXNIP mRNA and protein expression level and restrain the activation of NLRP3 inflammasome,reduce NAFLD model of rat liver inflammatory cytokines IL-1? and IL-18 level,thus improve the occurrence and development of NAFLD rat liver,for prevention and control of pomegranate peel polyphenols NAFLD certain theoretical basis.
