The branches with leaves of Viscum coloratum is a traditional Chinese medicine, named as Chinese Mistletoe in pharmacopoeia of PRC.Samples were collected from different origins and host trees.In the present study, the chemical constituents, the quality control methods and the metabolism of Viscum coloratum were investigated in detail.The chemical constituents of Viscum coloratum were systematically studied and 11 compounds were isolated and purified with silica gel, and sephadex LH-20 column chromatography. Their structures were identified as Lupeol acetate (E1),β-Sitosterol (E2), Oleanolic acid (E3),Pachypodol (E5), Syringaresinol (E6), Omuine (E7), Quercetin-3, 3’-dimethyl ether (E8),1,7-Di(4-hydroxybenzene)-heptane-1,4-diene-3-ketone (E9), Syringin (E10) and 2S-Hmoeriodictyol-7-O-β-D-glucoside (Ell).Compound E1 was isolated from Viscum coloratum for the first time. E5 and E7 were isolated from Viscum L.for the first time.To develop a high performance liquid chromatography (HPLC) method for the determination of Syringaresinol in Chinese Mistletoe, a Diamonsil C18 column (250×4.6 mm, 5μm)with a mobile phase consisting of acetonitrile-water (30:70, v/v) was used, and the detective wavelength was set at 272 nm. The Linear range of syringaresinol was 0.0174~0.348 mg·mL-1 (r=0.9995). The average recovery of Syringaresinol was 98.0% with RSD of 1.6%(n=9). A high performance liquid chromatography method was developed for the simultaneous determination of four Flavonoids in Chinese Mistletoe. The separation was performed on a Diamonsil C18 column (250 mm×4.6 mm,5μm) with the gradient elution solvent of acetonitrile and methanol-0.1% phosphoric acid (40:60, v/v). The detective wavelength was set at 280 nm. There were good linear relationships between the mass concentrations and the peak areas of hmoeriodictyol, quercetin-3,3’-dimethyl ether, omuine, and pachypodol in the range of 0.04 12~0.824 mg·mL-1(r=0.9994),0.00778~0.1556 mg·mL-1(r=0.9990),0.0360~0.720 mg·mL-1(r=0.9990),0.00320~0.0640 mg·mL-1(r= 0.9990),respectively.The recoveries of four flavonoids were 97.63%(RSD=1.6%),96.4%(RSD=1.9%),97.5%(RSD=1.7%),and 98.1%(RSD=2.5%),respectively.This method is sensitive and suitable for the quality control of Chinese Mistletoe.Homoeriodictyol-7-O-β-D-glucoside,possessing various pharmacological effects,was paid close attention to. After intravenous injection and intragastric administration of homoeriodictyol-7-O-β-D-glucoside,original glucoside,homoeriodictyol-7-O-β-D-glucoside glucuronide conjugates and homoeriodictyol glucuronide conjugates were detected in urine.
