Viscum coloratum(Komar.)Nakai is a dry foliated stem branch of mistletoe genus of the mulberry parasitic family.It has the functions of dispelling wind and dampness,nourishing liver and kidney,strengthening muscles and bones,placating fetal cells and clearing meridians.In this study,Viscum coloratum(Komar.)Nakai was taken as the research object.From the aspects of investigation of mistletoe current standard,detection of heavy metals,detection of forbidden agricultural residues,establishment of lilacin thin layer identification,optimization of oleanol thin layer identification,establishment of total alkaloid content determination method of mistletoe,establishment of HPLC multi-component content determination method,and establishment of mistletoe characteristic spectrum and mistletoe flavonoid chemical composition fingerprint,The existing standards were studied comprehensively to provide data support for more effective quality control of mistletoe.The specific research is as follows:1.The properties,microscopic,moisture,ash,extract and impurities of the collected mistletoe samples were determined,and the rationality of the existing standards was investigated.Results A total of 28 batches of samples were collected.S1-S26 were identified as mistletoe Viscum coloratum(Komar.)Nakai,S27 is Viscum articulatum Burm.f.,S28 is Taxillus sutchuenensis(Lecomte)Danser.Samples S1-S26 all conform to the current pharmacopoeia regulations.2.The TLC identification of syringin from Viscum coloratum(Komar.)Nakai was established.In TLC identification,the test product showed the same color spots in the corresponding position as the control drug of mistletoe and the control drug of syringin,which showed good specificity.The preparation method of test product in TLC identification of oleanolic acid was optimized.3.Through systematic liquid-mass analysis,the content determination indexes were screened and determined,the appropriate content determination indexes were determined,and the HPLC content determination method was established.A HPLC method was established to determine the contents of mistletoe samples in different batches,and the results were analyzed comprehensively.The established method can achieve good separation of each index component in the sample,and the methodological investigation conforms to the regulations.The content of Viscumneoside Ⅲ was between 2.02 and 6.91 mg/g,the content of Homoeriodictyol-7-O-β-D-Glycoside was between 0.88 and 2.93 mg/g,and the content of Homoeriodictyol was between 0.85 and 2.44mg/g.The determination of total alkaloids in Viscum coloratum(Komar.)Nakai was established by UV.The results showed that the content of total alkaloids in mistletoe samples was between 1.46 and 2.87 mg/g.Considering the contents of four components in the samples comprehensively,there were differences between different producing areas and different batches of samples from the same producing area,and the content of index components in leaves was higher than that in stems,indicating that different producing areas and different parts would affect the quality of Viscum coloratum(Komar.)Nakai.4.HPLC method was used to establish the full spectrum of Viscum coloratum(Komar.)Nakai.Eight common peaks were identified and two chromatographic peaks were identified.Combined with the analysis of chemical measurement method,the quality of Viscum coloratum(Komar.)Nakai was different among different producing areas and batches,and heterotropic compounds peak 1,peak 7,peak 6 were screened.The established atlas could distinguish Viscum articulatum Burm.f.and Taxillus sutchuenensis(Lecomte)Danser.HPLC method was used to establish the fingerprint of mistletoe flavonoids,and 7 common characteristic peaks with basically the same peak time and large peak area in the fingerprint of Viscum coloratum(Komar.)Nakai were determined for similarity calculation.The similarity of 20 batches of Viscum coloratum(Komar.)Nakai was greater than 0.9,and the methodology results showed that the established method was accurate and convenient.Through the systematic study of Viscum coloratum(Komar.)Nakai,the quality standard of it was improved,which provided a reference for the quality evaluation of it. |
