| Objective:To investigate the extraction and purification process of the saponins from Radix Astragali overground parts. To provide a reference for the development of the overground parts of Astragalus membranaceus. compare the overground parts with the underground parts of Astragalus membranaceus. The Radix Astragali overground parts studied based 1H NMR metabolomics approach.Methods:With the content of saponin of Radix Astragali overground parts as index,select the best extracting process for saponins from Radix Astragali overground parts. Several resins were compared in this paper to select the best purification process. The comparison of the difference between the overground parts and underground parts of Astragalus membranaceus was conducted in the aspects of chemical constitutions and pharmacological activities. 10 kinds of their chemical constituents were analyzed by muti-components determination, and their effects on marrow hematopoietic function in mice caused by cyclophosphamide damage were evaluated with blood counts as an index.A metabolomic study based on 1H NMR has been performed to analysis the constitutions of the overground parts and underground parts of Astragalus membranaceus. The Radix Astragali overground parts induced metabolic changes in the endogenous metabolites in mice were studied based 1H NMR metabolomics approach.Results:The optimal conditions were obtained as follows: the solvent is 80% ethanol, the extraeting time is 30 min and solid-liquid ration is 1:40. In order to select the best purifying process for saponins from Radix Astragali overground parts. Several resins were compared in this paper. And D101 macro porous resin was selected as the best column. The best purification process was as follows: the raw material was 0.1 g·mL-1. The extracts were adsorbed by D101 and eluted by 8 volume of water and 5 volume of 40% ethanol. Then the resin was eluted by 5 volume of 70% ethanol, and the absorption velocity of flow is 2 BV/h.We collected 70% ethanol eluant components. The process has good application prospects.The chemical composition in the overground and underground parts of Astragalus membranaceus were similar, but the content of astragaloside IV, astragaloside III, ononin,calycosin, formononetin, 7,2’-dihydroxy-3’,4’-dimethoxy isoflavans the overground parts were higher than those in the underground parts. The pharmacological effect of the two parts had no significant difference, which increase the number of white blood cells, red blood cells, hemoglobin of the damaged mice. They all had no apparent side effect.Metabolomics results show that alterations among the endogenous metabolites in the serum little difference between the overground and underground parts of Astragalus membranaceus except individual amino acids in the same does. The overground and underground parts of Astragalus membranaceus may have pharmacological consistency in some aspects.Conclusion:The extraction and purification process of saponins from Radix Astragali overground parts was determined, and the content of total saponins reached more than 50%. The chemical composition in the overground and underground parts of Astragalus membranaceus were similar. The pharmacological effect of the two parts had no significant difference, which increase the number of white blood cells, red blood cells, hemoglobin of the damaged mice without no apparent side effect. The overground and underground partsof Astragalus membranaceus were nearly similar from each other in regard to flavonoids and saponins as well as some primary metabolites. The changes of the above ground parts and the underground parts of Astragalus membranaceus were basically consistent with the changes of metabolism in mice. The overground parts of Astragalus membranaceus was valuable for further exploition. |
