Objective: Analysis of garlicin effects on human pancreatic cancer PANC-1 cell and molecular biology mechanism and further explore the effect of allicin on pancreatic cancer cells in vitro invasion metastasis, the application of allicin in the clinical treatment of pancreatic cancer to provide experimental and theoretical basis for.Methods: The PANC-1 Cells were cultured and treated with 10 ng·m L-1 TGF-β1ã€10 ng·m L-1 TGF-β1+5 μg·m L-1 Allicinã€5 μg·m L-1Allicin, respectively. Untreated cells were used as normal controls. Cell proliferation was detected by MTT method. The cell migration and invasion ability of pancreatic cancer cells was determined by scratch test and Transwell assay.The expression of E-Cadherin, Viminten and NF-κB in each group were detected by Real-time PCR.Result : MTT showed that allicin has inhibitory effect on pancreatic cancer cells in a dose and time-dependent manner;Scratch assay and Transwell show that allicin can significantly inhibit the migration and invasion of pancreatic cancer cells undergoing EMT. The difference was statistically significant(P < 0.05);Real time PCR showed that pancreatic cancer cells after allicin and TGF-β1treatment, compared with con group, TGF-β1 group of E-cadherin expression decreased significantly(P < 0.05), vimentin expression was significantly increased(P < 0.05) and that TGF-β1 to promote pancreatic cancer cells undergoing EMT.Compared with the TGF-β1 group, the expression of allicin +TGF-β1 group and Allicin group E-candherin up-regulated, vimentin, down regulated the expression of show that allicin has blocked TGF-β1 induced pancreatic cancer EMT effect; at the same time, allicin +TGF-β1and Allicin groups of the NF-κB expression also decreased significantly(P < 0.05), indicating that allicin can inhibit pancreatic cancer cell EMT through regulating the expression of NF-κB.Conclusion: allicin can inhibit the occurrence of pancreatic cancer EMT cells regulate NF-κB signaling pathway.Conclusion: 1.allicin can inhibit pancreatic cancer PANC-1 cells EMT.2. allicin can inhibit the occurrence of pancreatic cancer EMT cells regulate NF-κB signaling pathway. |
