Objective: In this study, fluorescent immunocytochemistry analysis of the positive expression rate of epithelial-mesenchymal transition related proteins(E-cadherin, Vimentin) and morphological changes in gastric cancer HGC-27 cells before and after allicin intervention.Initially by exploring the effects of allicin on gastric cancer cells EMT, which illustrating the effects of allicin on invasion and metastasis of gastric cancer cells in vitro.Methods: Gastric cancer HGC-27 cells to EMT by IL-6 stimulation;Before and after allicin intervention,Cells morphological changes were observed ueder inverted microscope,and the positive expression rate of EMT related proteins(E-cadherin, Vimentin)in gastric cancer cells was detected by fluorescent immunocytochemistry.Using SPSS17.0 statistical software to analyze the positive expression rate of these proteins before and after the intervention of allicin, as well as morphological changes of gastric cancer HGC-27 cells,To illustrate the effects of allicin on gastric cancer cells EMT.Result : 1.Morphological changes on gastric cancer HGC-27 cells:Negative control group, gastric cancer HGC-27 cells grow good, connection of intercellular is much and close, scattered cells is rare;In the positive control group,cells loss of the multilateral shape,intercellular adhesion is destroyed significantly, mostly dispersed isolated state, the majority of cells similar to fibrous, some cells showed a spindle, the performance characteristics of mesenchymal cells; In treatment group,intercellular adhesion of gastric HGC-27 cells increase,the state change Dispersion to close, gradually transform the mesenchymal cells characteristic into the epithelial cell properties after allicin intervention. 2. The positive expression rates of EMT related proteins:E-cadherin positive expression rate of the negative control group, positive control group and treatment group by fluorescent immunocytochemistry assay is 0.747 ± 0.031,0.360 ± 0.027,0.541 ± 0.024, Vimentin positive expression rates is 0.354 ± 0.019,0.685 ± 0.048,0.469 ± 0.035. Positive control group is stimulated by IL-6 gastric HGC-27 cells.Positive control group expressed a higher level of mesenchymal marker proteins(vimentin) but a lower level of epithelial marker proteins(E-cadherin),than Negative control group, and the difference was statistically significant(P <0.05); Treatment group is that Allicin cultured gastric cancer cells after IL-6 stimulation.Treatment groupexpressed a higher level of epithelial marker proteins(E-cadherin) but a lower level of mesenchymal marker proteins(vimentin),than positive control group,but do not reach the level of negative control group,and the difference was statistically significant(P <0.05).Conclusion: 1.allicin can inhibit gastric cancer HGC-27 cells EMT. 2. By allicin intervention the E-cadherin expression level of gastric cancer HGC-27 increase, while Vimentin expression levels down.
