Objective: To observe the expression of AQP5 and CC16 and the effect of Dahuangfuzi decoction on lung after the resuscitation of hemorrhagic shock in rats.Methods: Healthy and clean male SD rats(n = 16 per group) were randomly divided into three groups: sham operation(control group), or the resuscitation of hemorrhagic shock maintained for 1 h and then treated with either physiological saline enema(model group) or Dahuang Fuzi Decotion enema(treatment group). Besides model group and the treatment group were separately randomized to two subgroups included 12 h and 24 h(n = 8 per subgroup). The control group followed the line of intraperitoneal anesthesia,the right common carotid artery, the right femoral vein, the left femoral artery and systemic heparinization, without the resuscitation of hemorrhagic shock. The standard of recovery is the mean arterial pressure stability over 90-100 mm Hg. Based on preparing the model of the resuscitation of hemorrhagic shock, model group and treatment group were given physiological saline enema and 2ml Dahuang Fuzi Decotion enema on 12 h and 24 h, respectively. Each group abstracted blood of the rats to measure blood serum LPS, CC16 level before executing them according to different time points. Lung tissue was taken to measure lung water content(the ratio of left lung wet / dry weight) after the rats were executed; Damages of lung tissue were measured by HE staining; Examined the expression of CC16 and AQP5 in lung tissue by Immunohistochemical methods.Results: 1.The content of plasma lipopolysaccharide: model group was significantly higher than the control group(P <0.01). Comparing model group and treatment group, Plasma lipopolysaccharide levels in the treatment group was significantly reduced compared with model group, Both differences were statistically significant at all time points(P <0.05).2.The content of plasma Clara cell protein(CC16): the model group was higher than the control group(P <0.05). Comparing model group and treatment group, plasma CC16 content was higher in the model group than in the treatment group, this was statistically significant(P <0.05). 3. The content of pulmonary tissue homogenate AQP-5: compare with the control group,the model group was lower, this was statistically significant(P <0.05). But Compare with the model group, AQP-5 content was higher in the treatment group than in the model group(P <0.05). 4.Detection of rat lung tissue water content(the ratio of left lung wet / dry weight) : the model group and the treatment group were all higher than the control group(P < 0.05). lung tissue water content in the model group was higher than in the treatment group, Both differences were statistically significant at all time points(P <0.05). 5.Observe damages of lung tissue by light microscopy: the control group:alveolar structural integrity,alveolar sacs clear,alveolar septum normal.the model group:a large number of inflammatory cell infiltrates in alveoli and alveolar cell edema or bleeding, and alveolar septum broadening.the treatment group: inflammatory cell infiltrates in alveoli and alveolar cell edema or bleeding was significantly reduced compared with model group, there was significant difference between the two groups in lung injure score(P <0.05). 6.Lung tissue Clara cell protein(CC16) Immunohistochemistry: Positive staining: cytoplasm showing brown is rat lung Clara cell protein(CC16). Compare with model and treatment group, the score of control Group was significantly higher.The score of treatment group was higher than model group(P <0.05). 7. Lung tissue AQP5 Immunohistochemistry:Compare with model and treatment group, the score of control Group was significantly higher.The score of treatment group was higher than model group, the difference was statistically significant(P <0.05).Conclusion: The lung AQP5 and CC16 protein involved in acute lung injury process after the resuscitation of hemorrhagic shock in rats. Dahuangfuzi decoction can increase the lung tissue CC16 AQP5 protein and protein expression,and significantly reduce the degree of acute lung injury after hemorrhagic shock in rats,that was effective prevention and control of the progress of hemorrhagic shock.
