Respiratory Department, kunming general hospital, Kunming Medical College, 650032[Abstract] Objective: To investigate the effect of peritoneal injected glucocorticoids on the number of Clara cells and the expression of Clara cell protein 10 in lung tissue of a guinea pig model of asthma.Menthods : The 29 healthy guinea pigs are randomly divided into three groups: A normal control group, an allergic asthma group and a dexamethasone (DXM) pretreatment group, in wich a guinea pig asthma model was established by sensitization and challenge with ovalumin. Bronchial alveolar lavage fluid (BALF) were collected subsequently from the left lung of guinea pigs for examining the total cellnumber and cell classification. To investigate the number of Clara cells (CC10-positive cells) in lung tissue by immunohistochemical method and the pathological change of lung tissue in light microscope. Total RNA were isolated from the lung tissue of guinea pigs by onestep way , then it become cDNA by reverse transcription. Amplify the cDNA and measure its sequence. Results: The total cellular score and the number of eosinophile granulocyte of BALF of the allergic asthma group and DXM pretreatment group were increased than control group(P<0.05) . The number of Clara cells in lung tissue of the allergic asthma group were significantly decreased than that of control and DXM pretreatment groups (P<0.05) . Amplification product of cDNA , which was 793bp gene sequence. Conclusion : CC10 is an important endogenous anti-inflammatory factor, The decreasing the Clara cell number in lung tissue of the allergic asthma group suggest that the Clara cells and CC10 may participate in the pathogenesis of asthma. Glucocorticoids increased the expression of CC10 in lung tissue, and it may have an anti-inflammatory effect in the pathogenesy of asthma by increasing the CC10 levels of lung tissue. |
