Objective:Using FeCl3 induced posttraumatic epilepsy(PTE)model to observe llepcimide under the effect of different doses of epileptic seizure times and degree, effect of loss of neurons in the hippocampus of the neuroprotective effect of ilepcimide traumatic epilepsy. Methods:(1) Traumatic animal model: healthy adult male SD rats, in rats the left motor cortex injection of 5μL concentration is 0.2mol/L FeCl3(physiological saline control group except), according to criteria of Racine in 1972, seizures to grade 3 or 3 above for the successful modeling.(2) Experimental group: the rats were randomly divided into 5 groups, each group of ten, A: physiological saline control group received equal volume of saline gavage, 1 time/day for seven consecutive days. B: model control group, given equal volume of physiological saline gavage, 1 time/ day for seven consecutive days; C: llepcimide low dose group: rats given after llepcimide 200mg/kg intragastric administration for seven days; D: llepcimide dose group: after modeling llepcimide 400mg/kg intragastric administration for seven days; E: llepcimide high dose group: modeling llepcimide 800mg/kg intragastric administration for seven days.(3) Changes in behavior: the behavior of the rats after seven days of observation of drug intervention and the frequency of seizures.(4) Nissl staining: the neuronal loss in hippocampal CA3 region was observed.(5) Statistical analysis: the SPSS 17.0 statistical software was used to analyze the data of each group. The single factor analysis of variance was used to compare the difference between groups, and LSD test the difference of P<0.05 was statistically significant. Results:(1) The number of epileptic seizures: to llepcimide each dose group and model group compared with normal saline control group, the difference was statistically significant(P < 0.05); D and E group compared with model group had significant difference(P < 0.05), except for group C(P > 0.05); C, D, E group compare, the difference had statistical significance(P < 0.05).(2) Neuron loss situation: from llepcimide each dose group and model group compared with normal saline control group, the difference was statistically significant(P < 0.05); D and E group compared with model group had significant difference(P < 0.05), except for group C(P > 0.05); C, D, E group compare, the difference had statistical significance(P < 0.05).(3) From llepcimide in this model, the positive correlation between dose and effect. Conclusion:Llepcimide in traumatic model made epileptic attack, and reduced the number of inhibited the hippocampal neuron loss, suggesting that ilepcimide has neuroprotective effect.
