Objective To explore cerebral protective role of glucose-6-phosphatedehydrogenase(G6PD)in the early post-traumatic epilepsy. MethodsTwenty-four SD rats were randomly divided into three groups: controlgroups, experimental groups, intervention groups. The rats of interventiongroups were received an intraperitoneal injection of0.1μg/ml1,25-dihydroxyvitamin D3(1,25-(OH)2D3) for seven days,and the othersadministered with an steam-heated arachidonic acid. Rat models ofepilepsy were established by cerebral intracortex microinjection of100mmol/L ferric chloride solution5μl and identified byelectroencephalography (EEG) in2h later and occurrence of seizures bybehavior observation in6h later, and the control groups, administered withan intracortical injection of normal saline. Expression of G6PD mRNA andprotein in cerebral cortex were detected by RT-PCR and Western blot. Themalondialdehyde (MDA) level of lipid peroxidation and the antioxidantlevel of glutathione(GSH) and G6PD activity were detected by colorimetricmethod. Results Typical seizures were observed in experimental and intervention groups. The seriousness and duration of epilepsy seizures ofrats from intervention group were less obvious than those of model groups(P<0.05). The expressions of G6PD mRNA and protein in injection zoneof cortex from intervention groups were higher than those from the modelgroups (P<0.05), but lower than those from the control groups(P>0.05).The G6PD activity and GSH level increased (P<0.05) and the MDA levelsignificantly reduced in intervention groups (P<0.05).Conclusion G6PDcan improve the antioxidant capacity of brain cortex and protect rats fromthe FeCl3-induced epilepsy.
