Objectiveconstruct atherosclerotic animal model with gradually increase of plaque instability,through a high-fat diet, balloon injury, gene transfection technology,andresearch the relationship between the expression level of serum UII and plaque UII.Methodspurebred male Japanese white rabbits randomly divided into four groups,respectively group A(normal feed)〠group B(high-fat group), group C(high fat and balloon injury group), group D(high fat group plus balloon injury and gene transfection), raise 10 weeks, extract venous blood every week, and in the second week, carry out balloon injury and in the eight week,carry out vascular ultrasound and gene transfection surgery, put to death and obtain materials after 10 weeks.Though the application of sappanwood eosin staining, immunohistochemical staining,PCR method, observe the plaque formation and UII/UT expression distribution in the plague and quantitative m RNA, and study the relationship between serum UII level and UII m RNA.ResultsHE staining show, group A complete endothelial cells of right iliac artery, close to the elastic layer, neatly smooth muscle cells, group B partial or total loss of artery endothelial cells,visible atheromatous plaque, group C endothelial cells rupturecompletely, visible atheromatous plaque or fibrous plaque, a large number of inflammatory cells, group D similar to group C,in the aspect of right iliac artery, but the part of abdominal aortic we can see thinner fibrous cap, visible intraplaque neovasculature compared with group C.Immunohistochemical examination showed the color brown marked UII distribution is mainly distributed in the surrounding of macrophages and fibrous cap, and according to their group A and group B, group C,group D order,the color gradually deepened.B〠Cã€D groups plasma level of UII significant increase compared with group A,Cã€D groups increase compared with group B, group D the plasma levels of UII increased compared with group C, but there was no statistically significant difference(A 1.83±0.05pg/ml, B2.13±0.09pg/ml,C 3.09±0.17pg/ml,D 3.21±0.15pg/ml,Bã€Cã€D VS A,Cã€D VS B,P<0.05,C VS D,P>0.05). Group D after 1 week of P53 gene transfection(the 8th week, 3.5 days after transfection) plasma UII levels rise,compared with before transfection 1 week(7 weeks), but there was no statistically significant difference(3.49 ±0.21 pg/ml, 3.25 ±0.24 pg/ml, P > 0.05). Group D 2, 3 weeks after the P53 gene transfection(9, 10 weeks) plasma UII levels increase compared with gene transfection after 1 week(8 week), there was no statistically significant difference(3.62± 0.47 pg/ml, 3.72±0.39 pg/ml, 3.49± 0.21 pg/ml, P > 0.05). Group D P53 gene transfection plasma UII after 3 weeks a significant rise compared with the past transfection(3.59 ±0.27 pg/ml, 3.07 ±0.15 pg/ml, P = 0.004). After gene transfection 1 week(8 weeks) group D plasma UII level increased compared with group C, there was no statistically significant difference(3.49 ±0.21 pg/ml, 3.37±0.34pg/ml, P > 0.05).(4) the plaque area UIIm RNA expression, C 〠D groupssignificantly increase compared with Aã€B( group A 1.36 ±1.34, group B 4.88 ±1.80,group C 43.35±43.87,group D 41.91±35.52, P < 0.05).(5) the level of plasma UII and plaque UIIm RNA expression present positive correlation, r is 0.686, P = 0.000,the regression equation is Y(blood) = 2.636 + 0.015 X(plague).ConclusionsHigh fat feed, balloon injury, P53 gene transfection can successfully establish the rabbit model of atherosclerosis plaque formation and rupture of animals. Plasma levels of UII reflects the progress and stability of atherosclerotic plaque.The repair progress after atherosclerosis and plaque rupture, the UII and UT expression in atherosclerotic plaques increase, with the high level in plasma. |
