Background The pathogenesis of preeclampsia is still unknown. The physical vascularremoulding of spiral arterial decreases the resistance of the terminal vessels in placenta, and realizes the efficient placenta perfusion to be accordant of the development of pregnancy. Soluble fms-like tyrosine kinase-1 (sFlt-1), an inhibitor of vascular endothelial growth factor (VEGF) and placental growth factor (PLGF), is involved in the pathogenesis of preeclampsia.Objectives The study is aimed to detemine the relationship of sFlt-1 in the serum and placenta, the hypertensive disorder complicating pregnancy (HDCP), and the normal pregnancy.Methods 1 Twenty-two women with the HDCP which within six gestational hypertension, three mild preeclampsia, ten severe preeclampsia and three eclampsia were enrolled in this study. Eighteen healty pregnancies were recruited as the control group. Serum was obtained from the HDCP and the healthy control groups for analysis of the sFlt-1 concentration by using the enzyme linked immunosorbent assay (ELSIA).2 Twenty-three women with the HDCP, which within seven gestational hypertension, twelve preeclampsia, four eclampsia were enrolled in this study. Twenty healthy pregnancies were recruited as the control group. Placenta tissues were obtained from the HDCP and the healthy control groups for analysis of the sFlt-1 mRNA expression by using the quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). Results 1 The concentration of the sFlt-1 was higher in the HDCP group than in the control group (5.09±6.22ng/ml vs 1.02±1.12ng/ml, P<0.05). Furthermore, the concentrations of the sFlt-1 in the serum of severe preeclampsia were significantly higher levels 9.35±7.20ng/ml than that of the control group (P<0.01).2 The expression of the sFlt-1 mRNA were not different between the HDCP group and the control group (1.45±1.47 vs 1.04±0.69, P>0.05). While the expression of sFlt-1 mRNA in the placentas of eclampsia group were significantly higher levels 2.99±2.47 than that of the control group ( P<0.01).Conclusion 1 The concentration of the sFlt-1 in the serum of HDCP was higher than the normal pregnancy. Furthermore, the concentration of the sFlt-1 was significantly increased in women with severe preeclampsia. sFlt-1 may be contribute to the development of the HDCP .2 The results suggested that the sFlt-1 is necessary for pregnancy. The expression of the sFlt-1 was increased in women with eclmpsia.
