Objective:To examine thc effects of Prevention and therapy of herbal simple substanceRcsvcratrol on the cxperimcntal liver fibrosis(LF)of rats, and the differenceexpression of TGF-β1 which canonical correction with fibrosis in liver. Themechanism of this therapeutic effect of liver fibrosis will be discussed.Methods:The model of hepatic fibrosis in rats was induced by subcutaneously injecting 40%CCL4. and dimidiatcd into Preventing and therapy groups. There arc 6 teams bcPartition, normal, model, low dose, middlc dose, high dose and controlteam(colchicines) in each Prcvcnting and therapy group. Serumalanincaminotransfcrasc(ALT), thc ratio of albumin and globulin(A/G), hyaluronicacid (HA), hydroxyproline(Hyp), malondialdchydc(MDA), superroxide dismutase(SOD), typeⅢcollagen(pc-Ⅲ),α-smooth muscle action(α-SMA)wcre dctected.Morphological changes werc evaluated by Professional Pathologist.The exprcssionof transforming growth factorβ1 (TGFβ1) during the thcrapcutic Proccss of liverfibrosis were chccked by RT- PCR analysis.Results:Resveratrol could obviously improve the rats' mental state,appetite and hairluster, increase the their body wcight, decrease thc serum activity of ALT,raise theserum A/G, decrease thc amount of Hyp, MDA of the liver, increase the amount ofSOD,α-SMA,decrease the level of serum HA, pcⅢ. Serum ALT of all teamsgiven medication decreased markedly(P〈0.05), and the ratio of albumin andglobulin(A/G)increased markedly in high dose team. decreasing the level of serum ofHA, pc-Ⅲ, and decreasing Hyp content of liver tissue,There were marked differenceon HA,pc-Ⅲ, between model and high or low dose team of the therapy groups(P〈0.05).Morphological changes: the medication simple substance could effectivelyreduce the degree of liver fibrosis induced by CCL4. It could relieve the degenerationand necrosis of hepatocytes and reduce(all or Part)fibrous connective tissue deposited in the liver lobules, approaching to the normalliver tissue basically, there wereobvious difference between high dose and normal team (p〈0.01), but no significantdifference between the normal and control team(p〉0.05).Expression quantity of TGFβ1mRNA in normal team different notably from those in the high dose, low dose,normal and control team(P〈0.01)by RT-PCR in rat liver.Conclusion:Resveratrol has evidence in Preventing and repairing effects on the liver fibrosisinduced by 40% CCL4 and high dose group superior than other groups. Themechanism of this therapeutic effect of liver fibrosis correlates tightly with itsprotecting liver cells, adjusting metabolism of extracellular matrix (ECM). Relieveremarkably the inflammation damage, fatty degeneration of the liver cells, and reducethe collagen fibre of the liver.
