Good fetal health is the foundation of a healthy life. Medication during pregnancy is very important in fetal health. Chinese medicine is widely applied during pregnancy.Pregnancy toxicity of traditional Chinese medicine has also been more and more attention. Pinellia, which has good efficacy in dampness and phlegm, vomiting Jiangni, plays an important role in treatment of pregnancy vomiting, cough, dizziness. But in ancient literature, the pregnancy security applications of Pinellia is controversial. Modern experimental studies have shown that Pinellia toxicity pregnancy. Pinellia ginger ginseng compound is famous in the treatment of pregnancy Vomiting. What Pinellia pregnancy toxicity mechanism is? How does Ginseng ginger Pinellia compound detoxication? In this study,we detect the expression of pregnancy-associated protein in mice gastrulation with Western blotting after the using of Pinellia and ginseng ginger Pinellia compound,to explore the toxic effects of pregnancy-associated protein and factors that ginger ginseng Pinellia compound detoxification.ObjectiveThe Pinellia and Pinellia ginger ginseng compound, respectively acts to form a powder and decoction in mice during early pregnancy, to explore Pinellia pregnancy toxicity associated protein, and to study the impact of ginseng ginger Pinellia compound detoxification.MethodsPregnant mice were divided into four different groups. In the 6th day of pregnancy intragastric gavage for 3 days. Normal control group fed distilled water 0.4ml/day; Pinellia powder group fed 0.17g/ml Pinellia suspension 0.4ml/day; Pinellia decoction group fed 0.17g/ml Pinellia decoction 0.4ml/day; Pinellia compound powder group were fed 0.21g/ml ginger ginseng Pinellia suspension 0.8ml/day (in two times,eath 0.4ml), Pinellia compound decoction group were fed 0.43g/ml ginger ginseng Decoction 0.4ml/day.On the 6th day of gestation pregnant mice were sacrificed cervical off to detecting fetal tissue Fibrinogen β (FGB), tubulin (α-Tubulin), calmodulin (CaM) expression levels.Results1. Fibrinogen (FGB) expression:raw pinellia powder group, raw decoction group, compound powder group were lower than the normal control group (P<0.01). Compound decoction group and the normal control group,the difference was not statistically significant. Pinellia powder group and Pinellia decoction group showed no significant difference. Compound powder group was lower than compound decoction group (P<0.01). Pinellia powder group and compound powder group had no significant difference. Pinellia decoction group was lower than compound powder group (P<0.01).2. a-tubulin (a-Tubulin) expression:There was no statistically significant difference in each group and the normal control group. Pinellia powder group and Pinellia decoction group showed no significant difference. Compound powder group was lower than compound decoction group (P<0.05). Pinellia powder group and compound powder group had no significant difference. Pinellia decoction group was lower than compound powder group(P<0.05).3. calmodulin (CaM) expression:Pinellia powder group, Pinellia decoction group were lower than the normal control group(P<0.01). Compound powder group, compound decoction group were higher than the normal control group (P<0.01). Pinellia powder group and Pinellia decoction group showed no significant difference. Compound powder group, compound decoction group showed no significant difference. Pinellia powder group was lower than compound powder group (P<0.01). Pinellia decoction group was lower than compound decoction group (P<0.01).Conclusions1. Pinellia powder, Pinellia decoction, Pinellia ginger ginseng powder gastrulation in mice may inhibit the expression of fibrinogen; Pinellia ginger ginseng compound on mice gastrulation fibrinogen expression effects may have related to the compatibility and boiling.2. Pinellia powder, Pinellia decoction, Pinellia ginger ginseng powder, Pinellia ginger ginseng powder decoction could no significant effect on the expression of tubulin Mice gastrulation.3. Pinellia powder, Pinellia decoction in mice may inhibit the expression of Calmodulin; Pinellia ginger ginseng powder, Pinellia ginger ginseng decoction on mice gastrulation calmodulin expression may enhanced role; Pinellia ginger ginseng compound on mice gastrulation Calmodulin expression effects may have related to the compatibility.4. Pinellia toxic effects of pregnancy may inhibit the fibrinogen protein and fiber and calmodulin, leading to embryonic bleeding, bruising, cell division, differentiation, metabolic abnormalities; Pinellia ginger ginseng compound by boiling compatibility and can be lifted Banxia fiber inhibiting the expression of fibrinogen, compatibility can be lifted by Pinellia inhibiting Calmodulin expression, so as to achieve pregnancy solution Pinellia toxic effect.
