ObjectiveTo study the expression of C-erb B-2, P53, Ki67, Topo Ⅱα in breast cancer tissue, analyze the relationship between the expression and the clinicopathological factors, prognosis of breast cancer, and discuss the role of their expression of C-erb B-2, P53, Ki67, Topo Ⅱα in the occurrence, development and prognosis in breast cancer, in order to provide theoretical basis for gene target therapy for breast cancer.Methods82 patients with completed clinical data who were breast cancer pathologically confirmed after operation from January 2008 to December 2014 in Fengxian District Central Hospital were selected. 30 cases of normal colonic mucosa tissues at 3 cm outside the tumor edge were also selected as control group. The expression of C-erb B-2, P53, Ki67, Topo Ⅱα in the tissue of breast carcinoma and para-carcinoma tissue were detected by using immunohistochemical staining(S-P), the correlation between these indicators above and pathological features such as age, clinical stage, tumor diameter, pathological types and lymph node metastasis were analyzed. All the data was analyzed by SPSS 17.0 software, enumeration data was used Chi-squared test, Spearman rank correlation was used to analyze the relationship among C-erb B-2, P53, Ki67, Topo Ⅱα. Kaplan-Meier survival analysis were used to analyze the relationship between expression of C-erb B-2, P53, Ki67, Topo Ⅱα and time to progression(TTP). P<0.05 was considered statistically significant.Results1. The expression of C-erb B-2, P53, Ki67, Topo Ⅱ α in breast carcinoma tissue was 48.8%, 53.7%, 69.5%, 59.8%, and 10.0%, 6.7%, 10.0%, 13.3% in para-carcinoma tissue, there was statistically significant(χ2=13.97, 20.04, 31.28, 18.99, P<0.05).2. The positive expression rate of C-erb B-2 was correlated with pathological type, tumor diameter, clinical stage, lymph node metastasis(P<0.05). The positive expression rate of P53 was correlated with the clinical stage, lymph node metastasis(P<0.05). The positive expression rate of Ki67 was correlated with tumor diameter, tumor diameter, lymph node metastasis(P<0.05). The positive expression rate of Topo Ⅱ α was correlated with the clinical stage, tumor diameter, lymph node metastasis(P<0.05).3. The expression of C-erb B-2 was positively correlated with the expression of P53, Topo Ⅱ α in breast carcinoma(r=0.349, 0.694, P<0.05), and P53 was positively correlated with the expression of Ki67, Topo Ⅱ α in breast carcinoma(r=0.316, 0.436, P<0.05)4. The rate of postoperative local recurrence in C-erb B-2, P53, Ki67, Topo Ⅱ α negative was 26.2%, 26.3%, 24.0%, 27.3%, which was significantly lower than those in C-erb B-2, P53, Ki67, Topo Ⅱα positive 60.0%, 56.8%, 50.9%, 53.1%(P<0.05).5. Kaplan-Meier survival analysis showed that median TTP of all patients was 33.4 months(95% CI: 24.8~42.2). The median TTPs in positive C-erb B-2, P53, Topo Ⅱα expression group were significantly longer than those in negative expression group(P<0.05). The median TTP in positive and negative Ki67 expression group was not statistically significant(P>0.05).Conclusion1. The expression of C-erb B-2, P53, Ki67, TopoⅡα in breast cancer tissue are obviously higher than those of para-carcinoma tissue, and is closely correlated with pathological characteristics such as diameter, clinical stage, lymph node metastasis, detection of the expression of C-erb B-2, P53, Ki67, Topo Ⅱ α has important reference value in the judge of lesion properties, development trend, formulating reasonable treatment scheme.2. The expression of C-erb B-2, P53, Ki67, Topo Ⅱ α in breast cancer has significant postive correlation, and has influence on postoperative recurrence and survival time, all taking part in the occurrence, development and metastasis of breast cancer, which is expected to become the predictors of prognosis and potential therapeutic targets. |
