Gastrointestinal dysfunction is a common complication after abdominal surgery, and its clinical manifestation in the early stage could be disappearance of gastrointestinal of peristalsis, no gurgling sound, ileus and abdominal heaviness etc, which affects digestion. As a result, it is against postoperative wound healing. Abdominal distension may result in postoperative wound dehiscence or upper shift in diaphragm and causing dyspnea as tharocic is oppressed. All of them are life threatening. Then it is obvious that effective intervention for the postoperative gastrointestinal dysfunction in the early stage, is of great significance for rehabilitation after surgery.ObjectiveBy studying the influence of the 3-leg points in Jin’s 3-Needle technique on postoperative gastrointestinal activity adjustment, which is determined through the expression of c-kit and iNOS positive cells in colon sample.MethodColonic anastomosis is done to rats to make gastrointestinal dysmotility model, detailed as:narcotize rats by intraperitoneal injection of 10% chloral hydrate and incise abdomen along the mid-line about 2cm, then suture in situ after cutting 1/3 colon laterally 2cm away from colon, after which peritoneum and muscle are sutured continuously, and the abdominal skin is sutured discontinuously. All rats are divided into normal control group, model group and acupuncture group whose acupuncture treatment begins in the first day after model is made. The acupuncture group after modeling the first day acupuncture consecutive 10 days. Model group only modeled operation without acupuncture intervention. Nothing is done to control group. Lavage 1 ml ink per rat and kill them after 30 minutes to both acupuncture and the corresponding model group when acupuncture is finished. Measure the rate of small intestine propulsion, examine the expression of c-kit protein by western blot method and also the expression of iNOS positive cells by IHC, all of which are obtained from lcm colon sample where anastomosis is carried out.Results(1) Defecating weight:Model Group is lower than Acupuncture Group and Blank Group (P<0.01), but there is no statistical significance between Acupuncture Group and Blank Group 3(P>0.05)(2) The propulsive rate of small intestine:Compared with blank control group, Model Group is lower in propulsive rate(P<0.01), but there is no statistical significance between Acupuncture Group and blank Group (P>0.05).(3)Results of c-kit protein expression:Compared with blank control group, Model Group is lower in propulsive rate(P<0.01), but there is no statistical significance between Acupuncture Group and blank Group (P>0.05).(4) The result of iNOS IHC:Compared with blank control group, Model Group is lower in propulsive rate(P<0.01), Compared with the model group, the acupuncture is lower, the difference was statistically significant (P<0.01).Conclusions1. Acupuncture can effectively improve the gastrointestinal dynamics of intestinal anastomosis rats and promote gastrointestinal function recovery.2. (1)Intestinal anastomosis reduces the c-kit protein of cajal interstitial cells in intestinal tissue, causing gastric bowel function disorder. (2)Acupuncture promote the expression of the c-kit protein significantly.3. Acupuncture promoting cajal cell expression of c-kit might resume its microenvironment repair inhibition of iNOS expression.
