Primary liver cancer the second only next to lung cancer of all the cancer is on e of the world’s most common cancer. The purpose of this research was to study the inhibitory effect and its mechanism of Rolfe polysaccharides and Achyranthes bidenta ta polysaccharides on H22 liver cancer cell.Kunming mice were inoculated with H22 cells and H22 liver ascites type model is set up with 400 mg/kg 0.2ml polysaccharide lavage per mice for 20 days. The living and survival conditions in mice.were recorded to establish a solid tumor H22 liver cancer model. Each group had 10 mice and the groups including normal group, model group, mountain arrowhead polysaccharide high, medium and low dose group, cyclophosphamide group, observe and record 100、200、400mg/kg 0.2ml polysaccharide influence on solid tumor liver cancer in mice, quality records, test groups of mice body weight, tumor, inhibitory rate, spleen and thymus index. ELIS A was used to detect the expression of IL-2 and TNF alpha.in the Pleione yunnanensis Rolfe polysaccharides and Achyranthes bidentata polysaccharides on mice solid tumor H22 liver cancer after the first analysis of experimental results, the next batch experiment was carried out, the purpose was to explore the Pleione yunnanensis Rolfe polysaccharides best antitumor dose. At the same time increased the contrast Achyranthes bidentata polysaccharides. Solid tumor H22 liver cancer model is set up, each group of 10, only 10 group and normal group, model group, mountain arrowhead much sugar low, middle, high, high dose group, cyclophosphamide group, observe and record 300,600,900,1200 mg/kg Pleione yunnanensis Rolfe polysaccharides influence on solid tumor liver cancer in mice, Achyranthes bidentata polysaccharides.more 400,800,1200 mg/kg three dose groups, record Pleione yunnanensis Rolfe polysaccharides, Achyranthes bidentata polysaccharides, inhibitory rate and quality of each weight, tumor in mice spleen and thymus index, the ELISA detection of IL-2 and TNF alpha expression, HE staining, cell morphology, Immunohistochemical SABC method to detect the BCL-2, and the effects of P53 protein expression to explore the Pleione yunnanensis Rolfe polysaccharides and Achyranthes bidentata polysaccharides Induction of apoptosis mechanism.Results:Pleione yunnanensis Rolfe polysaccharide on the life extension of H22 ascites type mice is16.36%. Compared with model group,Has a very significant different (P<0.05). Pleione yunnanensis Rolfe polysaccharide to 100 mg/kg,200 mg/kg and 400 mg/kg in mice liver cancer H22 solid tumor inhibitory rate respectively 21.58%、32.76%、40.05%. High dose group of mice spleen index has the extremely significant (P< 0.01). The high, medium and low dose group of thymus index compared with model group and has significant difference (P< 0.01).300,600,900,1200 mg/kg, Pleione yunnanensis Rolfe polysaccharide inhibitory rate reached 43.74% in the 300 mg/kg.Achyranthes bidentata polysaccharides 400, 800,1200 mg/kg.in 400 mg/kg showed good antitumor and inhibitory rate reached 58.82%.Through experiment shows that the polysaccharide doses group were not significant side effects, the spleen index (P< 0.05), compared with model group. Pleione yunnanensis and Rolfe polysaccharide can increase the content of IL-2 a tumor-burdened mice so as to improve the immunity of mice,300,600,900 mg/kg dose group is significant (P< 0.05). Achyranthes bidentata polysaccharides 800,1200 mg/kg dose group with extreme difference (P< 0.01). The two Polysaccharides on mice H22 solid tumor can be observed by HE staining can promote tumor cell fragmentation. And it can inhibit in different degrees of antiapoptotic factor expression of Bcl- 2, p53 expression increased with decreased tumor cell differentiation.Conclusion:Pleione yunnane-nsis Rolfe polysaccharides and Achyranthes bidentata polysaccharides had antitumor effect.
