Qian Jinba is the dry root of Flemingia philippinensis Merr.et Rolfe. It is usually used for the treatment of traumatic injury, chronic nephritis, bronchitis, women's vaginal discharge embolism and so on by Yao doctors. This paper studied its chemical composition and anti-tumor activities systematically.The chemical constituents of petroleum ether fraction,chloroform fraction and ethyl acetate fraction of Flemingia philippinensis Merr.et Rolfe. were analysed by modern chromatography. Seventeen compounds and one mixture were isolated. Their structures were elucidated by spectroscopic analysis and comparison with published data. The structures of seventeen compounds had been identified to be Lupeol(1), Eriosematin (2),β-Sitosterol(3), Flemichapparin C(6),3',4'-Dihydroxy-trans-cinamic acid myricyl ester(7), Flemiphilippinin A(8),Diisobutyl phthalate(9), Physcion(10), Genistein(11), Betulinic acid(12), Osajin(13),Octacosyl-3,5-dihydroxy-cinnamate(14), Lupinalbin A(15),2',4',5,7-Tetrahydroxyisoflavone(16),Wedelolactone(17),4',7-Dihy-droxyisoflavone(18),Erythroglaucin(19).Mixture was identified to beβ-Sitosterol (3),Stigmasterol(4) and Campesterol(5). Compounds 5,7,9,14,17,18,19 were isolated and identificated from the Flemingia genus for the first time;Compound 4 was isolated from this species for the first time.The anti-tumor activity of Compound 2,6,7,8,10,11,13,14,15 were studied by MTT method. The results showed that:5 flavonoids (Compound 2,8,11,13,15) had significant anti-tumor activity and a certain dose-effect relationship, but other compounds did not.5 flavonoids showed a certain degree of selectivity in the anti-tumor research.Osajin and Flemiphilippinin A displayed a good activity in the inhibition of MCF-7 breast cancer cells and IC50 values reached to 19.23±0.71μmol/ L and 15.93±1.93μmol/L.Lupinalbin A had apparent effect on the growth of cancer cell of MCF-7 and HepG2 and IC50 values reached to 22.21±1.69μmol/L and 27.06±3.66μmol/L.Genistein inhibitted the growth of cervical cancer Hela was outstanding. IC50 value reached to 22.41±5.62μmol/L.Volatile oils were exacted by steam distillation (SD) method, ultrasound-assisted extraction and steam distillation (UESD) method and CO2 supercritical fluid extraction (SFE-CO2) method from Yao medicine Flemingia philippinensis Merr.et Rolfe. The chemical composition and the antitumor activity on human lung cancer cells A549 were analyzed and compared to track with the chemical constituents of antitumor activity. The results showed that:For different extracted methods, the chemical composition and biological activity are quite different. The oil yield obtained by SD was 0.03%,by UESD was 0.08% and by CO2-SEF was 0.5%. The main components of volatile oils extracted by SD and UESD were terpenoids, but the main components extracted by CO2-SEF were alcohols and acids. At the volatile oil mass concentration of 150ug/mL, the inhibition rate of volatile oil exacted by SD on tumor cell A549 reached 69.13%, by UESD achieved 59.91%, but the inhibitory rate of volatile oil exacted by CO2-SEF only reached to 24.08%. Through a comprehensive comparative analysis showed that the main active ingredients inhibiting A549 lung cancer cells may be Himachalene and longifolene-(V4) in the volatile oil.This finding has not been reported so far.
