Changium smyrnioides Wolff (Mingdangshen), a kind of umbelliferae plant mainly being produced in Jiangsu, Zhejiang and Anhui provinces, is a valuable medicine in versions of Chinese Pharmacopoeias. Its root is used as a medicine starting from Ming and Qing Dynasties. The medicine can moisturize lung, dissipate phlegm to stop coughing, regulate stomach to stop vomit, detoxificaion and subside swelling. The root, stem and fruit of this plant have been reported at home and abroad, however, the study of the root bark from it has not been reported. This study aimed to research on the active constituents and pharmacological effect. The research results are as follows:1. The study of chemical constituents from the root bark of Changium smyrnioides Wolff: Using column chromatography and heavy crystallization methods to obtain the compounds from the supercritical fluid extraction of the root bark of Changium smyrnioides Wolff.9 compounds were identified and structurally elucidated by their physic-chemical properties and spectral analysis. They were pentacosanoic acid, heptacosanol, β-sitosterol, stigmasterol, isoimperatorm, Imperatorin, xanthotoxol, phellopterin and 5-hydroxyl-8-methoxy-psoralen.2. The antitumor activity of five furanocoumarins:(1) Anti-tumor effect in vitro experiments showed that:Five furanocoumarins had differently inhibition to seven tumors,i.e., SMMC-7721, HepG2, A-549,MKN-45, HeLa, MCF-7 and MDA-MB-231.IC50 was from 0.30 μg·mL-1 to 17.23 μg·mL-1. Especially isoimperatorin was most significant effective.IC50 was from 0.39 μg·mL-1 to 4.11 μg·mL-1. (2) Anti-tumor effect in vivo experiments showed that: Isoimperatorm had significant antitumor effect on mouse liver cancer and the inhibition rate was 66.38%. The inhibition rate of the positive drug was 66.93%. Isoimperatorm had protective effect on the immune organs while the positive drug could damage to it.3. Experimental study on the antihyperlipidemic and antioxidant effect of the root bark from Changium smyrnioides Wolff:(1) Experimental study on the antihyperlipidemic effect showed that:The outcomes of TC, TG, LDL-C and Al were deceased apparently, and HDL-C increased apparently in middle-dose group (500 mg·kg-1) and high-dose group (1000 mg·kg-1). At the same time, the ethanol extracts can increase the production of SOD, GSH-PX, CAT and T-AOC, and decrease the content of MDA and NO. It is suggested that,the mechanism may be related to their effects of lipid oxidation and oxygen free radicals scavenging. Besides, they all had protective effect on rat liver. (2) Experimental study on the antioxidant effect showed that:Five furanocoumarins can increase the viability of injured endothelial cells and imperatorin was the best. Imperatorin can increase the cell survival by 22%.4. Determination of different parts from Changium smyrnioides Wolff:Simultaneous determination of imperatorin, phellopterin and 5-hydroxyl-8-methoxy-psoralen in different parts of Radix Changii. The result showed that:The highest content was the root bark of Radix Changii,17.3 times higher than processed. Besides, the contents of cultivated Radix Changii were significantly higher than that of wild products.
