Experime ntal Objective:By analyzing the change of the zinc metabolism in obese rat models and the change of related enzymes, this experiment aims to provide a theoretical foundation for prevention and treatment of obese patients zinc metabolic disorders on basis of the expression level of the change of zinc and obesity related m RNA in the obese rats. Methods:20 male rats of average weight are randomly divided into high fat diet group(n=10) in which the rats are fed with high fat forage and normal diet group(n=10) in which the rats are fed with the basic forage. The rats in both groups are fed for 8 weeks and then they will be killed after the eyeball blood is taken. The following will be tested in the serum test: serum iron, superoxide dismutase(sod) and angiotensin conversion enzyme activity, 5 `- nucleotide enzyme activity and the activity of alkaline phosphatase. The expression level of metallothionein m RNA will be tested from the livers; The expression level of ZIP4 and Zn T1 m RN A will be tested from the small intestines; the expression level of leptin m RNA will be tested from the adipose tissue. All theses above tests are upon timing and quantitative PCR methods. Results:1. The indicators of serum Zn, superoxide dismutase(sod) and angiotensin conversion enzyme activity, 5 `- NT and alkaline phosphatase rise in the obese rat group, which have a statistical significance compared with the control group(P < 0.05).2. Compared with control group, liver metallothionein m RNA expression has increased in obese rats, and the difference is statistically significant(P < 0.05).3. Compared with control group, the small intestine ZIP4 m RNA expression is reduced of the obese rats, and the difference is statistically significant(P < 0.05); Zn T1 m RNA expression increased, and the difference is statistically significant(P < 0.05).4. Compared with control group, leptin m RN A expression in obese rats fat has increased, and the difference is statistically significant(P < 0.05). Conclusion:1. Obesity will cause the body zinc level changes, the indicators of serum Zn, superoxide dismutase(sod) and angiotensin conversion enzyme activity, 5 `- NT and alkaline phosphatase increased, and result in zinc metabolic increased.2. The metallothionein gene expression has increased in obese rats’ livers.3. The ZIP4 gene expression in obese rats’ small intestine has declined and Zn T1 gene expression has risen.4. The leptin gene expression in obese rats’ fat tissue has risen. |
