Objective: To clone mouse ZnT3 cDNA fragment and to determine the level of ZnT3 mRNA expression in mouse tissue. Method: A certain fragment of ZnT3 cDNA with the base sequence confirmed by dideoxynucleotide method was cloned by RT-PCR. The expression of ZnT3 mRNA was examined by RT-PCR and its expression level was compared in mouse tissue with (-actin as control. Result: A single fragment about 700bp, its sequence conformable to corresponding fragment sequence of reported ZnT3 cDNA, was obtained. ZnT3 mRNA was detected in cerebrum, hippocampus and testis(especially higher level in testis),and not detected in heart, liver, lung, spleen, kidney, intestine , olfactory bulb and cerebellum. Conclusion: The proper fragment of ZnT3 cDNA has been cloned. The expression of ZnT3 mRNAin brain and testis suggests the possible significance of ZnT3 in brain and reproduction function.
