| ObjectiveTo study the effect of Edsysonein (EDS) on rabbits chondrocytes injury induced by lipopolysaccharide (LPS).MethodsAricular chondrocytes were isolated from rabbits, and randomly divided into three groups:control group; LPS group, chondrocytes with LPS induced injury; LPS+CS group, injury chondrocytes treated with chondroitin sulfate (CS); and LPS+EDS group, injury chondrocytes treated with EDS. The cell proliferation and cell apoptosis of chondrocytes were determined by MTT method and flow cytometry assay, respectively. The mRNA and protein expression level of inducible nitric oxide synthase (iNOS) were detected by RT-PCR and western blot. In addition, the content of NO and IL-1β were measured by nitric acid reductase method and enzyme-linked immunosorbent assay (ELISA) method, respectively.ResultsThe proliferation was decreased, cell apoptosis was increased, and the expression level of iNOS and NO and IL-1β contents were increased in model group of chondrocytes injury, which were significantly reversed by CS or EDS treatment (100μmol/L) (p<0.05).ConclusionThe EDSysone exhibited a protective effect on LPS induced rabbits chondrocytes injury through inhibiting the expression of iNOS. The effect is equall to CS. |
