Objective:Applying isobaric tags for relative and absolute quantitation combined liquidchromatography tandem mass spectrometer techniques to screen the differentially expressed serum proteins between patients with Chronic Hepa titis B belongs to liver-stagnation and spleen-deficiency syndrome and Spleen-St omach dampness-heat syndrome, provide a basis for objective research and Diff erential Treatment of TCM syndromes of chronic hepatitis B.Methods:In this study, according to previous clinical and literature researc h done by the research group, we choose both outpatients and inpatients that in line with Western medicine, Chinese medicine diagnostic criteria and exclusi-on criteria of chronic hepatitis B and two TCM syndromes in West China Ho-spital of Sichuan University with healthy volunteers from Chengdu, as the res-earch objects. We collected the objects venous blood,then centrifuged them to get the serum used for screening serum differentially expressed serum protein s by using iTRAQ labeling and LC-MS/MS then accomplished their biological analysis.Results:A total of 61 serum proteins were identified between liver-stagnat ion and spleen-deficiency syndrome and Spleen-Stomach dampness-heat syndro me. Of the 61 differentially expressed serum proteins,16 were up-regulated an d 45 down-regulated. These differentially expressed proteins are involved in 35 biological processes,16 cellular components,26 molecular function, and 6 path ways. YWHAH, RhoC, RhoA, ATP5A1, ATP5B and CDHlwere found to be located in the connected nodes of the protein functional interaction network.Conclusion:Due to our research, differentially expressed serum protein spe ctrum was found. YWHAH, RhoC, RhoA,ATP5Al,ATP5B and CDH1 located in the connected nodes of the protein functional interaction network may be th e biological bases of differences between two TCM syndromes.
