| Objective:To observe the effect of HIFU ablating osteosarcoma enhanced by Phase-change type Poly lactic-co-glycolic acid microspheres (Pct-PLGAM) and the influence of Pct-PLGAM with different concentrations on the enhancing efficacy, in order to investigate the feasibility of Pct-PLGAM enhancing efficacy of HIFU ablating the osteosarcomaMethods:Pct-PLGAM was prepared and detect its form, particle size, surface potential and the condition of Phase-change were detected. Xenograft tumor model of human osteosarcoma were established for as the experimental objective and divided into blank group, control group and experimental group(treated with the indicated concentration of Pct-PLGAM emulsion) randomly, then each group was subjected to the HIFU ablation immediately. Specimens were taken to H&E staining and necrosis area of osteosarcoma were calculated and compared between each group at 0 h and 72 h after HIFU ablation. Proliferation and apoptosis changes of the targeted region were analyzed after 72 h-HIFU ablation.Results:There were significant differences in the area of necrosis of osteosarcoma after the HIFU ablation between the control group and experimental groups with high concentration of Pct-PLGAM(P<0.05),but not with experimental group with low concentration of Pct-PLGAM(P> 0.05).Experimental groups with high concentration of Pct-PLGAM had a lager area of necrosis after 72 h-HIFU ablation(P<0.05),other groups had not.There were significant differences in apoptotic index and expressing rate of PCNA out of target regions between the control group and experimental groups(P<0.05).Conclusion:In the Xenograft osteosarcoma model,the Pct-PLGAM can enhance the efficacy of HIFU ablation osteosarcoma,and the effect rely on the concentration of Pct-PLGAM. |
