| Purpose:To explore the radiotherapy sensitization on radiation of Aged Black Garlic Extract (ABGE) on lung cancer cell Line lewis and colon cancer Ht-29 in mouse and preliminary discuss the possible molecular mechanism. Methods:Inhibition rate of lung cancer was detected by MTT. Effect of colony formation was observed by Cloning formation assay. Changes of cell morphology was observed by Hoechst staining. Flow cytometry used to detected changes in the cell cycle. The expressions of Bcl-2 and Bax of lewis cells treated with different concentrations of aged black garlic extract were detected with Western blot. The tumor-bearing C57BL/6 mouse modle was established, 40 mice were randomized into 4 groups:Control group(peritoneal injection 0.9%NS,0.2 ml):γ-ray group(peritoneal injection 0.9%NS,0.2 ml+γ ray,2Gy/day):Aged black garlic extract(ABGE) group(peritoneal injection, 1g/ml aged black garlic extract 0.2 ml):Aged black garlic extract+γ-ray group(ABGE+γ-ray group)(peritoneal injection, 1g/ml aged black garlic extract 0.2 ml+γ ray,2Gy/day). The tumor volume and tumor growth delay (TGD) were observed, the tumors were separated and weighed to calculate the tumor-inhibiting rate. Flow cytometry Annexin V-FITC/PI was used to detect tumor cell apoptosis. Realtime-PCR was used to detect tumor Bcl-2, Bax, Caspase-9 protein expression. The amounts of WBC in peripheral blood, the superoxide dismutase (SOD), catalase(CAT) activity were measured.Results:(1) ABGE could had significantly inhibitory action on growth of lewis lung cancer cell. The combination of aged black garlic extract and radiotherapy significantly induced lewis cell apoptosis and affected cell cycle,upregulating the expression of Bax and downregulating the expression of Bcl-2. (2) ABGE combined with radiotherapy could significantly increase the tumor-inhibiting rate, experience a delay in tumor growth and improve the radiotherapy sensitization enhancement ratio. Aged black garlic extract could promote tumor cell apoptosis, downregulate the expression of Bcl-2 and upregulate the expression of Bax, Caspase-9 mRNA, Aged black garlic extract could increased the amounts of WBC and SOD,CAT activity in mice.Conclusion:(1) ABGE could sensitize the lung cancer lewis to ionizing irradiation. This effect is probably cauesd by increasing the rate of G2/M cells or upregulating the expression of Bax and downregulating the expression of Bcl-2.(2) ABGE could sensitize colon cancer Ht-29 xenografts to enhance radiotherapy by down-regulating the expression of Bcl-2 and upregulating the expression Bax and Caspase-9. Aged black garlic extract also showed a protective effect on irradiation injury in mice by increasing the amounts of WBC in peripheral blood and improving the antioxidant ability of mice. |
