Currently a variety of combination therapy for patients with lung cancer improve survival rate, however, the overall survival and life quality of lung cancer patients is far from people’s expectations. Especially, The traditional chemotherapy drugs, which do harm for patients with cancer, often lead patients to refuse treatment. Chinese medicines which used to treat tumor have many advantages, such as low toxic side effects, improved symptoms significantly, increased life quality and low cost in long-term treatment of patients with cancer. Therefore, the study of Chinese medicines on tumor treatment is payed attention by more and more scholars.The modern pharmacology and clinical applications show that Chamaejasme can inhibit tumor cell proliferation, prevent cell division and induce apoptosis effect, with a broad spectrum anti-tumor and a variety of anti-tumor components. However, there are different ideas about antitumor components of Chamaejasme, some people think that the anti-tumor ingredient is water-soluble components, another think that it is fat-soluble ingredients. Considering the interaction between Chamaejasme lipophilic components and water-soluble components, based on folk method in which the Chinese medicine for cancer treatment is boiled with a half of wine and a half of water, we extract Chamaejasme with50%ethanol. We think that the extraction from50%ethanol can retain the water-soluble and fat soluble active ingredients, anti-tumor effect of the extracts was studied in vitro and in vivo and its implied mechanism was explored.In vitro, MTT assay was used to test the effect of ethanol extract from Chamaejasmeon (EEC) on cell proliferation in Lewis lung cancer cells, cell motility was investigated by cell adhesion assay, wound healing assay and transwell migration assay; cell autophagy was examined by acridine orange staining. while it was also measured the levels of cellular glucose (Glu), glucose transporter1(GLUT1), triglyceride (TG) and free fatty acids (FFA),and Using western blot detection EEC on affect of autophagy protein LC3and Beclinl gene. In vivo determination, the inhibition rate of EEC with Lewis lung cancer subcutaneous allograft,while the levels of GLUT1, lactate dehydrogenase (LDH), succinate dehydrogenase (SDH), TG and FFA in tumor tissues were detected; while the effects of EEC on tumor size and metastasis were evaluated with Lewis lung cancer experimental lung metastasis.the GLUT1and fatty acid synthase (FAS) in lung region with tumor or not were compared by immunohistochemistry.EEC suppressed cell attachment and proliferation as well as cell motive in a dose dependent matter in Lewis lung cancer cells in vitro, it could decrease the levels of cellular Glu, GLUT1, TG and FFA. and promote the autophagy in Lewis lung cancer cells.In vivo result, compared with untreated model mice, EEC reduces tumor size and metastasis and prolongated life span in a dose dependent matter. Under this experimental conditions, The inhibitiory rate of EEC is43.46%at High-dose and39.58%at Low-dose.respectively.EEC was able to decrease the levels of GLUT1,SDH, TG and FFA, but promoted LDH activity in tumor tissues.Western blot experiments showed that EEC can significantly increase autophagy Beclinl and LC3. In immunohistochemistry, EEC could inhibit GLUT1and FAS in lung region with tumor.In summary, EEC has a clear therapeutic and anti-metastatic effect on the Lewis lung cancer, and its mechanism may be related to the regulation of cell energy metabolism. |
