A Study Of Youguiwan’s Molecular Mechanism On Intervening Senior Citizens With Kidney Yang Deficiency From Related Genetic Research Of Immunity

Objective:By the experiment of fluorescence quantitative PCR, to testing the changes of immune genes quantitative expressions before the intervene by Youguiwan of Kidney yang deficiency patients, to explain part of the Molecular mechanism of the intervene of Youguiwan of kidney yang deficiency syndrome, to provide scientific basis for the prevention of aging kidney yang deficiency.Methods:1. Combing through ancient and modern literature, Summarizes the physiological function of kidney Yang and the common clinical symptoms of kidney Yang deficiency syndrome;Research Team summarized the effect of common diseases which are treated by Youguiwan.2. To research the aging kidney Yang deficiency and healthy controls whole-genome microarray datum which are clustered analysis by3DPCA, getting the aging kidney Yang deficiency syndrome differences in gene expression profile, combining the biological functions of genetic analysis, deleting selected differentially expressed genes in the immune class.3.People who bring into the aging kidney yang deficiency constitution, will be given Youguiwan to treat for28days, and we will evaluate efficacy by half a quantitative diagnostic criteria of Kidney yang deficiency.4.Testing the difference of the immune genes quantitative expression level of aging kidney Yang deficiency before and after used Youguiwan by RT-PCR.Results:1. Therapeutic principles and prescription of the Youguiwan fits in with the deficiency of kidney yang pathogenesis, which has a good effect on a variety of diseases such as andrology、gynecology、orthopedics department、internal secretion and brain, there is a wider range of clinical applications.2. Based on early differential gene expression profiles of deficiency of kidney yang, we screened28immune genes for testing genetic quantitative expression differences before and after the intervention of Youguiwan.3. We incorporated17cases of aging kidney yang deficiency, there are15patients who have the clinical manifestations of the kidney yang deficiency had improved significantly through the Youguiwan intervention, the total efficiency is88.2%.4. There is one gene NCOA2has a significance of statistical obviousness of Youguiwan on kidney yang before and after the intervention, and six genes FHIT、COL13A1、IRAK2、SSTR5、TIAM2、PTGER1have a significant difference.Conclusion:1.
