Objective: Taking the patients of Kidney-yang deficiency with genual Osteoarthritis as studing samples, to analyse its differential gene expression profile and molecule pathway and so to further explore the bio-molecular background of Kidney Yang -deficiency.Method: Using of cDNA microarray technology, basing on the studing background of accumulated symptom scores of Kidney-yang deficiency and bio-molecular results of genual Osteoarthritis, we select two groups of tapical patients who have been fulfillly diagnosed by professors of TCM and orthopedics, at the same time control group is also been selected, every group includes of 8 peoples. They are all evaluated as follows: â‘ Evaluateing the autonomic nerve quotiety. â‘¡Gene differtential expression profiling:the procedure of gene chip experiment include: drawing Blood, extracting mRNA, labeling, hybridizing, scanning, data analyzing. At last the data of two differently expressed genes between patients group and control group are obtained.Results: The diagnostic results of selected patients:the Kidney-yang deficiency scores of two groups of patients are all exceed 18,contrasted to control group ,there is a significant difference in the analyse of statistics. Combined with the diagnosis of the professor of the orthopedics and detection by X-rays, they are all tipical patients who satisfied with our standard of patient's selection. â‘¡ The results of the evaluating on autonomic nerve quotiety: two groups of data of autonomic nerve quotiety all reach no statistics difference. â‘¢Differential gene expression profile: 75 differential expression genes are obtained in chip 1, in it 49 are up-regulated and 26 are down-regulated; 1302 differential expression genes are also obtained in chip 2, and there are 291 up-regulated and 1011 down-regulated;6 genes in both two chips are also obtained.
