Objective: To study the correlation between the ex Pression of IL-4、IL-5、 IFN-γ m RNA and its methylation level in Patients with allergic rhinitis(AR),and Provide a basis for the diagnosis and treatment of AR.Methods: 50 Patients diagnosed with acute AR in our hos Pital from January 2016 to January 2017 were selected as ex Perimental grou P.Meanwhile,50 healthy Peo Ple in the Physical examination center were chosen as the control grou P.Peri Pheral blood was collected from all enrolled subjects.The ex Pressions of IL-5,IL-4 and IFN-γ m RNA were detected by Real Time PCR,and the Protein ex Pressions of IL-5,IL-4 and IFN-γ were detected by ELISA.The degree of methylation of IL-5,IL-4,and IFN-γ were detected by Na Pshot.Then,the differences of gene ex Pression,Protein ex Pression,and methylation of IL-5,IL-4,and IFN-γ were com Pared between the two grou Ps..Results:1.Com Parison of the m RNA ex Pression level of IFN-γ,IL-5 and IL-4 m RNA in the two grou PsCom Pared with the control grou P,the ex Pression of IFN-γ m RNA in the ex Perimental grou P was significantly lower(P<0.05),whereas the ex Pressions of IL-5 and IL-4 m RNA were statistically higher(P< 0.05).2.Com Parison of the Protein ex Pression level of IL-5,IL-4,and IFN-γ in two grou Ps.Com Pared with the control grou P,the IFN-γ Protein ex Pression was statistically significant lower in the ex Perimental grou P(P<0.05).However,the IL-4 and IL-5 Protein ex Pressions in the ex Perimental grou P were higher than those in the control grou P(P<0.05).3.Com Parison of the methylation levels of IL-5,IL-4 and IFN-γ between two grou PsCom Pared with the control grou P,the methylation level of IFN-γ in Promoter site was significantly higher in the ex Perimental grou P(P<0.05),and as for the IL-5 and IL-4,was lower than that of in the control grou P(P < 0.05).4.Correlation between the ex Pressions of the three cytokines and their methylation levels in the ex Perimental grou PThe m RNA ex Pression of IL-5,was negatively correlated with its degree of methylation level(r=-0.542,P= 0.000),so were IL-4(r=-0.796,P=0.014)and IFN-γ(r=-0.796,P=0.014).Conclusion1.The m RNA and Protein ex Pressions of IL-5 and IL-4 in AR Patients were increased,while the ex Pression of IFN-γ was decreased.2.The ex Pression of IL-4、IL-5、 IFN-γ m RNA is closely related to the level of Promoter methylation in AR Patients.The ex Pression of IL-4,IL-5,IFN-γ m RNA was negatively correlated with the degree of methylation.
