| Objective To investigate the expressions of serum thymidine kinase 1(S-TK1) in patients with laryngeal cancer and healthy people,and research the correlations between the concentration of S-TK1 in laryngeal carcinoma histological grade, TNM staging and S-TK1 expressions before and after operation. To further explore the significance of the change of expressions of S-TK1 before and after the operation to assess the curative effect of operation.Methods 103 cases of laryngeal cancer patients were collected from the department of otolaryngology head and neck surgery, the First Affiliated Hospital of Medical University Of Anhui during 2013 April to 2014 October. All patients with laryngeal carcinoma were pathologically diagnosed as squamous cell carcinoma, and all patients were treated by surgery. All patients in the group of laryngeal carcinoma before operation, one week after operation and one month after opreation were stay to take morning fasting blood samples. In addition, to collect 129 cases of healthy persons blood specimens from the physical examination center of the First Affiliated Hospital of Medical University Of Anhui. Use enhanced chemiluminescence dot blot assay to detect the concentration of serum TK1 of laryngeal cancer group and control group, and record the S-TK1 concentration of each experimental subject and the number of positive and negative cases. Data were analyzed by SPSS17.0 software, the difference between the groups of measurement data using two independent samples t test or t test of paired, count data using c2 test and average number of pairwise comparisons between groups using analysis of variance. P <0.05 was considered as statistically significant..Results 1.The S-TK1 concentration of laryngeal cancer group were higher than that of control group[(2.95±2.99)pmol/L vs.(1.18±0.68)pmol/L](P<0.05). No significant correlations were found between laryngeal cancer group before operation and laryngeal cancer group after seven days of operation [(2.95±2.99)pmol/L vs.(2.82±2.31) pmol/L](P>0.05).But S-TK1 levels were higher in laryngeal cancer group before operation than those in laryngeal cancer group after one month of operation[(2.95±2.99)pmol/L vs.(1.72±1.30)pmol/L](P<0.05). 2.According to TK1 concentration higher than 2 pmol/L as positive diagnostic criteria, the positive rate of S-TK1 in laryngeal carcinoma preoperative was 49.51%, which significantly higher than the positive rate of 12.40% in the control group(P<0.05). A week after the surgery of laryngeal cancer group, the positive rate of S-TK1 was 54.37%, compared with the preoperative had no significant difference(P>0.05). However, one month after operation, the positive rate of S-TK1 was 26.21%, significantly lower than that of preoperative and one week after operation(P<0.05). 3.The level of S-TK1 in low differentiation was higher than that of high differentiation, but there is no associated with laryngeal cancer TNM staging. 4.The sensitivity and specificity of S-TK1 diagnosis of laryngeal cancer were 49.51%(51/103) and 87.59%(113/129).Conclusion 1.S-TK1 expression level were significantly elevated in patients with laryngeal cancer,while low expression in healthy people. The results suggest that S-TK1 can be used as auxiliary indexes of laryngeal cancer diagnosis and health screening. 2.There is a certain correlation between the expression of S-TK1 in laryngeal carcinoma patients and laryngeal histological grading,so we can see the relevant of S-TK1 concentration and the degree of tumor malignant. 3.The S-TK1 levels in laryngeal cancer group after one month of operation werelower than those in laryngeal cancer group before operation, suggesting that there is a certain significance to evaluate and detect the operation effect. |
