objective:This research is mainly by immunohistochemical analysis of Colorectal Carcinoma(CRC)tissue pathological slices, detect Pyruvate kinase M1(M1-PK) and Pyruvate kinase M2(M2-PK) in different clinical and pathological staging of CRC organization and expression in normal colorectal tissues adjacen t to carcinoma. In order to obtain the relationship with the clinicopathological ch aracteristics of CRC with the result, exploring the M1-PK and M2-PK in CRC expression with the relationship between disease development.Methods:This exper iment specimens from the pathology department of the first people’s hospital of Yi Bin, In January 2013- January 2015 detection of CRC tissue samples of 69, corresponding to the normal colorectal tissues adjacent to carcinoma specimens of 69, all the pathological specimens are complete with first class medical records of clinical diagnosis.Medical records, specification in detail, the operation data a nd image information is complete.Patient treatment process in detail,Specimen sou rce procedures detailed in patients aged 45-82, the median age is 65, the male 32 cases, female 37 cases. There are 20 cases of papillary adenocarcinoma, 41 ca ses of tubular adenocarcinoma and 8 cases of other pathological types carcinoma in the histologic types of CRC,The type of CRC pathology contains 48 cases of middle differentiation carcinoma and 21 cases of low differentiation car cinoma.Using immunohistochemical streptavidin-peroxidase(SP) immunohistochemi cal staining,Detect M1-PK and M2-PK in CRC tissues and normal colorectal tiss ue expression of differences,comparing the low differentiated group, the middle differentiated group with the normal tissue adjacent to carcinoma.Using single fac tor and multiple factors analysis comparing M1-PK and M2-PK in CRC low dif ferentiated group, the middle differentiation in the group with normal tissue adjac ent to carcinoma in the relationship between the expression and clinical pathologi cal features.Using correlation analysis and correlation analysis to detect CRC used to arrange express the correlation between M1-PK and M2-PK group.Results:(1)The expression and distribution in colorectal tissue of M1-PK and M2-PK: M1-P K and M2-PK have been expressed in the normal colonic epithelium cells and CRC epithelial cells.Arranged in the cytoplasm distribution, positive signals chara cterized by tan particles.M1-PK expression in CRC tissue, lower than that of nor mal tissue adjacent to carcinoma(Average optical density,AOD: 0.320±0.026 i n the control group; the cancer group 0.231±0.023 P < 0.05);M2-PK expression in CRC tissue, however, significantly higher than that of normal tissue adjacent t o carcinoma(AOD: 0.207±0.056 in the control group; the cancer group 0.347±0.054 P < 0.05).The expression of M1-PK in CRC tissues decreased as the degree of tumor differentiation(AOD: 0.320 ± 0.026 in the control group; middle differ entiated group 0.260 ±0.016; low differentiated group 0.167 ± 0.059; P < 0.05);The expression of M2-PK in CRC tissues increased with tumor differentiation de gree reduced(AOD: 0.207±0.056 in the control group; middle differentiated grou p 0.259±0.068; low differentiated group 0.420 ±0.019; P < 0.05);(2). Using singl e factor and multiple factors analysis and comparison of M1-PK and M2-PK in CRC low differentiated group, the middle differentiation in the group with norma l tissue adjacent to carcinoma in the relationship between the expression and clin ical pathological features show that the positive expression of M1-PK in CRC tis sues is closely related to the degree of differentiation(P < 0. 05), but with the patient age, tumor size, tumor location, clinical TNM stages and lymph node me tastasis. M2-PK in the tissue of CRC positive expression and the tumor size, tu mor is closely related to the clinical TNM staging and differentiation degree of t umor(P < 0. 05), but with the patient age, tumor location, presence of lymph n ode metastasis, no obvious relationship.(3). M1-PK in CRC organization the expr ession level of relevance to the M2-PK expression: using the linear correlation a nalysis results show that the M1-PK compared with M2-PK express AOD, r = 0.689, P < 0.01, suggesting a negative correlation relationship.Conclusions:(1)The re are M1-PK and M2-PK expressed in CRC epithelial cells and normal colonic epithelium, and specifically expressed not only one of them.(2)M2-PK expressio n in CRC cells initial response tumor size, tumor stage and tumor differentiation degree.(3) From the perspective of organization to express that the M2-PK with malignant degree and the progress of human CRC were positively correlated, b ut M1-PK with people negatively correlated with the malignant degree and the p rogress of CRC. |
