The Influence Of Xing Nao Jie Yu Capsule On PSD Model Rat’s Hippocampus ERK1/2

Objective: To establish post-stroke depression(post- stroke ’, PSD) model in rats, observe intervention effects of Xing Nao Jie Yu Capsule on on PSD model rat`s brain hippocampus p-ERK1/2 and hippocampus p- ERK1/2m RNA, and to explore the mechanism of Xing Nao Jie Yu Capsule in the treatment of PSD from the cellular and molecular levels, then to provide reliable experimental basis for the more extensive use in clinical treatment of PSD with Xing Nao Jie Yu Capsule.Method:1.Using the MCAO to design a permanent and focal Cerebral ischemia model,on the base of Cerebral ischemia model,combine the way to cure separatly and using Compound Reserpine Tablets to desigh the rat model of PSD.2.Selecting 120 healthy SD rats,a half of male and a half female,whose weight are 200±20g.According to The Desert Trails score,the rats were classified 8 groups at a random:Blank control group,Blank TCM group, Blank westerm-medicine group,Model group,TCM early intervention group, Western-medicine early intervention group,TCM group,Western-medicine group.3.Feeding adaptively after a week,all groups were desighed the Cerebral ischemia model except for Blank control group,Blank TCM group, Blank westerm-medicine group,on the base of Cerebral ischemia model,combine the way to cure separately and using Compound Reserpine Tablets(for 16 days)to desigh the rat model of PSD.After successful Cerebral ischemia model was desighed,from the second day the rats of Blank TCM group,Blank western-medicine group,TCM early intervention group,Western-medicine early intervention group were filled the stomach for 37 days;for PSD model,the rats of TCM group and Western-medicine were filled the stomach for 16 days.Using Xinnaojieyu Capsule 0.4125 g /kg to dissolved in 2 ml of distilled water to fill the stomach for all TCM groups;using FIuoxetine hydrochloride 2.08 mg /kg and Aspirin Enterie Ccoated Tablets 3.0mg/kg for all western-medicine groups.4.Acording to mearuring weight,sugar-water consumption and score of Open Field Test in 2th day,15 thday,37th day to obseve Xingnaojiahas an impact on the indicators above,of rats suffering from Strok with depression.5.Using the immunohistochemeic to analyse the content of the p-ERK1/2 outside the acidifical cells in the all group of rats, hippocampus. 6.Using the RT-PCR to test Xingnaojieyu Capsule has an influnence on the p-ERK1/2m RNA espression in the PSD rats, hippocampus.Result:1.Xingnaojieyu Capsule had an influnence on the weight and behavior of PSD rats.After desighing the model,compared with Blank control group,TCM control group and Western-medicine group,the weight,sugar-consumption was obviously redused,so did the horizontal motion and vertical movement of rats.After treatment,compared with Model group,the indicators above were obviously higher,the most obvious was TCM early intervention group and Western-medicine early intervention group.The result showed Xingnaojieyu Capsule can make the weight,sugar-consumption of PSD model rats increased,and improve the indicators of behavior.2.Mearuring the content of p-ERK1/2 in the hippocampus of ratsCompared with Blank control group,the p-ERK1/2 cells that arranged loosely and positive expression of shallow color in the hippocampus of rats were obviously reduced,which had an significant difference and statistical significance(P<0.01).However,compared with the Blank control group,positive cells reduced(P<0.05).Compared TCM early intervention group with TCM group,and compared TCM group with Western-medicine group to reduce the positive cells,there were no significant difference(P>0.05).3.Mearuring p-ERK1/2mRNA espression of in the hippocampus of all kinds of rats.Compared with Blank control group,the p-ERK1/2m RNA espression of in the hippocampus of rats in Model group was reduced,which had an statistical significance(P < 0.01).Compared with the Model group,the p-ERK1/2m RNA espression of in the hippocampus of rats in the TCM early intervention group,TCM group and Western-medicine group increased in different degrees,there was statistical significance(P<0.01).Compared TCM group and TCM early invention group,TCM group and Western-medicine group,there were no significant difference(P >0.05).Conclusion:1.MCAO,curing separately,combine Compound Reserpine Tablets with building model can succeed in desighing the PSD model of rats.2.PSD model rats had some sympoms:the colour dark flatness,eat less,weight loss,lazy to move,slow activity,less sugur-consumption.Most of the sympoms which are improved after filled the Xingnaojieyu Capsule in the stomach,are same with the sympoms and pathological changes of PSD patients.3.Xingnaojieyu Capsule can improve the content of p-ERK1/2 in the hippocampus of rats and the level of p-ERK1/2m RNA espression,which showed protecting the neurons damage caused by stress and promoting nerve cell regeneration is one of the treatment mechanism of Xingnaojieyu Capsule.
