Objective:To observe the effects of XingNaoJieYu Capsule on rats’brain hippocampus CaMmRNA and CaMKⅡ of PSD model,by establish model rats of Post-stroke depression(PSD),and to explore its molecular mechanism of XingNaoJieYu Capsule in the treatment of PSD,which provides experimental basis for clinical treatment of PSD with XingNaoJieYu Capsule.Methods:Screening 120 SD rats with half males and half females,which were randomly divided into 8 groups according to the behavioral test results,control group,control of Chinese medicine group,control group of western medicine,model group,early Chinese medicine intervention group,early western medicine intervention group,Chinese medicine treatment group and western medicine treatment group.After one week of adaptive feeding,all groups except control group,control of Chinese medicine group,and control group of western medicine were given permanent focal cerebral is chemic model with the method of MCAO,and then given isolated feeding method and intraperitoneal injection of reserpine(for 14 consecutive days)based on focal cerebral is chemic model.After cerebral stroke models were successfully prepared,rats were given a gavage of medicine in control group of Chinese medicine,control group of western medicine,early Chinese medicine intervention group and early western medicine intervention group for 37 days.After PSD models were successfully prepared,gavage the rats in Chinese medicine treatment group and western medicine treatment group for 21 days.According to their weight,gavage the rats in Chinese medicine group by0.4125g/kg Xingnao Jieyu capsule dissolved in 2 ml of distilled water while gavage the rats in western medicine group by 2.08mg/kg Prozac and 3.0 mg/kg Baia aspirin dissolved in 2 ml of distilled water.Measure their weight,sugar consumption and open box test scores of rats respectively on the day of the second,the 15th and the 37th to observe the effect of Xingnao Jieyu Capsule on the above indexes.Use RT-PCR method to detect the effect of Xingnao Jieyu Capsule on CaMmRNA level in hippocampus of rats,and use western blot method to detect the content of CaMKⅡ expression in hippocampus of rats.Results:1.The effect of Xing nao Jieyu Capsule on the weight and behavior of PSD rats:after modeling,comparing the results with control group,control group of Chinese medicine and control group of western medicine,the neurological symptoms of rats were in different degree.Weight,sugar consumption decreased significantly,meanwhile,the level of exercise rats and vertical movement scores also significantly decreased.After medicine treatment,compared with model group,the above indexes of other groups significantly increased.Furthermore,the early intervention groups of both Chinese medicine and Western medicine affected most obviously,which showed Xingnao Jieyu Capsule could increase PSD rats’weight,sugar consumption and the vertical movement and horizontal movement,and promote the recovery of nerve function.2.Measure the contents of CaMmRNA in the hippocampus of rats.Compared with the control control group,the relative expression content of CaMKⅡ in hippocampus of rats in model group obviously reduced,which had an significant difference and statistical significance(P<0.01).However,compare with the model group,the relative expression content of CaMKⅡ in hippocampus of rats in early Chinese medicine intervention group,early western medicine intervention group,Chinese medicine treatment group and western medicine treatment group all increased in different degrees,which had an significant difference and statistical significance(P<0.01,P<0.05).Comparing early intervention group with Chinese medicine treatment group,and early western medicine intervention group with western medicine treatment group,they were both no significant difference(P>0.05).3.Measure CaMKⅡ expression in the hippocampus of all kinds of rats.Comparing with the blank control group,CaMKⅡ expression in the hippocampus of rats in model group reduced,which had an statistical significance(P<0.01).Compared with the model group,the CaMKⅡ expression contents in the hippocampus of rats in early Chinese medicine intervention group,early western medicine intervention group,Chinese medicine treatment group and western medicine treatment group were all increased in different degrees,which had an significant difference and statistical significance(P<0.01).Compared early intervention group with Chinese medicine treatment group,and compared early western medicine intervention group with western medicine treatment group,they were both no significant difference(P>0.05).Conclusion:1.If MCAO is cured separately,intraperitoneal injection reserpine with established model can succeed in designing the PSD model of rats.2.PSD model rats have some symptoms,such as weight loss,lazy movement,slow activity,less sugar-consumption,and different degrees of neurological damaging.Most of the symptoms are obviously improved after filled their stomaches with Xingnaojieyu Capsule,which are similar to the symptoms of PSD patients.3.Xingnaojieyu Capsule can improve the expression contents of CaMmRNA and CaMKⅡ in the hippocampus of rats,which shows protecting the neurons damage caused by stress and promoting nerve cell regeneration is one of the mechanisms of Xingnaojieyu Capsule to cure PSD.
