Objective This thesis is to study the invasive force of the folliclestimulating hormone (FSH) on epithelial ovarian cancer cell lines SKOV3,HO8910PM and HO8910with different concentrations and observe the expressionof E-catenin protein in these ovarian cancer cell lines after treated by FSH withdifferent concentrations so as to explore the relationship between FSH and theovarian cancer cells Wnt/E-catenin.Methods Ovarian cancer cell lines SKOV3, HO8910PM and HO8910weretreated by FSH with different concentrations of0,20,40,60and80mIu/ml for48hours. The cell lines which were treated by FSH with concentration of0composeda control group. Transwell experiment was used to measure the invasive force ofthese cell lines. And immunohistochemistry and Real time PCR were used todetect the expressions of E-catenin protein and E-catenin mRNA in these celllines.Results The invasive force of the positive FSH receptor in HO8910andHO8910PM was significantly enhanced (P <0.05) when they had been treated byFSH with concentration of40and60mIu.mL-1. And the expressions of theE-catenin protein and mRNA in the cell lines were significantly decreased (P<0.05). There was no apparent change (P>0.05) in the invasive force of thenegative receptor SKOV3at different concentrations of FSH. And the expressionsof E-catenin and mRNA in SKOV3had no statistical significance (P>0.05). Theexpression of E-catenin is detected in SKOV3, HO8910PM and HO8910. And the positive expression exists within the cytoplasm. After stimulated by FSH withdifferent concentrations for a period of48hours, the E-catenin expression wasdecreased (P<0.05) in HO8910PM and HO8910with concentrations of40and60mIu/ml. There was no statistical significance (P>0.05) in the E-catenin expressionin SKOV3after stimulated by FSH with different concentrations.Conclusion FSH could enhance the invasive force of ovarian cancer cellsHO8910PM and HO8910, with the best stimulating FSH concentrations40and60mIu/ml. FSH could not stimulate the invasive force of the FSH negative receptorSKOV3with and different concentrations. There were differences in the E-cateninexpressions in HO8910PM and HO8910after treated by FSH with differentconcentrations. There was no difference in the expression of E-catenin in SKOV3.Therefore, the invasion of FSH on the ovarian cancer cells might be achievedthrough Wnt to reduce the expression of E-catenin in the epithelial ovarian cancercell lines HO8910PM and HO8910.
