OBJECTIVE Euphorbia kansui as traditional Chinese medicine has importantclinical medicinal value and development prospects,but its toxicity and irritating greatlylimits the clinical application of euphorbia kansui. Liquorice is a traditional Chinesemedicine to relieve the drug,s toxicity and irritant. Kansui and Licorice is Eighteenimcompatible Pairs. So it is a very meaningful topic whether Kansui and Licoricecompatibility can relieve the Kansui’s toxicity. The traditional Chinese medicine theory asthe guide,this study is based on metabonomics method of nuclear magnetic resonance(NMR). And it is combined with traditional toxicology indicator analysis,and integration ofdata integration methods. Then it is using the theory of systems biology to study thedynamic change law of metabolic processes of biological fluids and tissues of rats that ithas eat Kansui Licorice compatibility. Through the metabolic response and tissue damage,we can clarify Licorice can relieve Kansui. METHODS1H-NMR and patternrecognition analysis were given Kansui,Kansui1:1compatibility of licorice and Kansui1:4of licorice liquid in rats. RESULTS After that,comparted with controls,each grouphad remarkable different in endogenous metabolites. Compared with the controls,themetabolites variation including (1)urine from rats treated with Kansui had higher levels ofalanine,lactate,trimethylamine oxide (TMAO),betaine,taurine,glucose,phenylalanineand hippurate,and lower levels of α-ketoglutaric acid,succinate,citrate,glycine andcreatine/creatinine;(2)plasma from rats treated with Kansui had higher levels of lactate,alanine,glycoprotein,creatine/creatinine,pyruvate,and lower levers of lipids,HB,taurine,unsaturated fatty acid and glycerol;(3)feces from rats treated with Kansui had higher levels of isoleucine,leucine,valine,alanine,propionate, acetate,glutamate,lysine,cadaverine,tyrosine and phenylalanine,and lower levers of bile acid,succinate,taurineand glucose;(4)liver from rats treated with Kansui had higher levels of isoleucine,leucine,valine,lysine,glutamine,choline,phosphorylcholine,glycerophosphorylcholine,tyrosine and phenylalanine,and lower levers of taurine,glycine,glucose and glycogen;(5)kidney from rats treated with Kansui had higher levels of isoleucine,leucine,valine,alanine,lysine,acetate,methionine,creatine/creatinine and phenylalanine,and lower leversof glutamine, choline, phosphorylcholine and taurine. These differences verifyhistopathological examination and serum biochemical analysis.We can make the conclusionthan Kansui Glycyrrhizae1:4can relicve Kansui’s toxicity. CONCLUSION Combiningthe toxicology and biochemistry knowledge,We found that the compatibility of Kansui4times the amount of licorice,which can relieve the body metabolic disorders (such as theKrebs cycle,sugar metabolism,energy metabolism,amino acid metabolism,lipid and fattyacid metabolism disorders,etc.) and intestinal dysbacteriosis.This study by monitoring andcomparing Kansui compatibility of Licorice group and Kansui group changes betweenmetabolites, to understand the mechanism of Licorice can reduce the toxicity of Kansui.Itcan provides a new method to study Chinese Compatibility.
