ObjectiveTo.research on the effects of chronic toxicity of Shizaotang Decoction and Shizaotang Decoction mixed with licorice on rat cardiac and pulmonary function and histomorphology, so as to improve Shizaotang Decoction and "Eighteen anti-" theory, to explore the compatibility mechanism of "toxic Chinese medicine" and "instead of drugs", and provide the scientific the experimental data for the clinical medication safety, promote the Chinese medicine theory of continuous improvement and development.MethodsSPF level 90 SD rats, half male and half female, were randomly divided into 7 groups: normal control group, high-dose groups of Shizaotang Decoction, medium-dose groups of Shizaotang Decoction, low-dose groups of Shizaotang Decoction, high-dose groups of Shizaotang Decoction mixed with licorice, medium-dose groups of Shizaotang Decoction mixed with licorice, low-dose groups of Shizaotang Decoction mixed with licorice, normal control group 12, the rest of the group with 13 rats in each group, normal feeding into the cage.1 weeks after adaptive feeding, rats with equal volume of unequal concentration of gastric perfusion, according to the weight of each administration, 100g rats were 2ml, the concentration of the high, medium and low-dose group of Shizaotang Decoction and Shizaotang Decoction mixed with licorice were respectively 0.28g,0.14g and 0.07g/(kg·d). The normal control group were given the same volume of distilled water, irrigation and continuous stomach 6 days a week,1 days off, a total of 90 days. Ninety-first days of withdrawal, the last 1 times after dosing fasted for 24 hours, each group were randomly selected half of rats after anesthesia, abdominal aortic blood test biochemical index of blood, liver, stomach and gross morphology was observed with the naked eye, the changes of liver, stomach tissue structure were observed under light microscope, the remaining half animal observed after discontinuation of 2 week, repeat the above check. In the course of the experiment the rats were observed and recorded daily appearance signs (such as hair color, skin, etc.), behavior, glandular secretion (eye, ear, nose, genital), excreta (color, characters, etc.) changes in diet, weigh 1 times per day 2-3, and adjusted according to the weight of intragastric volume.The analysis of the experimental data by using SPASS19.0 statistical software, P<0.05 had statistical significance.Results1 WeightThe experimental results showed that, before administration, in administration period, the rats body weight showed no significant difference (P>0.05). In recovery period, rat’s weight of Shizaotang Decoction grew faster than the others(P<0.05).2 Cardiac CoefficientsThe experimental results showed that, in administration period, the rat’s cardiac coefficients of each group showed no significant difference (P>0.05). Compared with the administration period, each group showed a great reduction(P<0.05).3 Myocardial Enzymes(AST,CK,CK-MB,LDH, a-HBDH)3.1 ASTThe experimental results showed that, in administration period, the rat’s AST of administration group had an obvious increase(P<0.05)compared with the control group. In recovery period, the rat’s AST of Shizaotang Decoction group decreased(P<0.05). AST of Shizaotang Decoction mixed with licorice group were still greater than that of the control group(P<0.05).3.2CKThe experimental results showed that, in administration period, the rat’s CK of administration group had an obvious increase(P<0.05)compared with the control group. In recovery period, the rat’s CK of Shizaotang Decoction group decreased (P<0.05). CK of Shizaotang Decoction mixed with licorice group were still greater than that of the control group(P<0.05).3.3CK-MBThe experimental results showed that, in administration period, the rat’s CK-MB of administration group had an obvious increase (P<0.05)compared with the control group. In recovery period, the rat’s CK-MB of the two medium-dose groups increased compared with the two group in the administration period (P<0.05). While, the CK-MB of the two high-dose groups decreased compared with the two groups in the administration periodCP<0.05).3.4LDHThe experimental results showed that, in administration period, the rat’s LDH of high-dose group had an obvious increase(P<0.05)compared with the control group, while other group showed no statistical significance(P>0.05). In recovery period, the rat’s LDH of administration group decreased(P<0.05), and LDH of the high-dose group was still above on the control group(.P<0.05). And this group in recovery period showed no statistical significance compared with the group in the administration period.3.5a-HBDHThe experimental results showed that, in administration period, the rat’s LDH of administration group had an obvious increase(P<0.05)compared with the control group. In recovery period, the rat’s LDH of Shizaotang Decoction group decreased(P<0.05). LDH of Shizaotang Decoction mixed with licorice group were still-greater than that of the control group(P<0.05).4 Cardiac HistomorphologyCompared with control group, the rat of each administration groups showed some kind of heart tissue damage, with myocardial cells slight edema, myocardial fibers disorder loose, RBC leaking, and flake inflammatory infiltrates. The damage degree is closely related to medicine dose.5 Pulmonary CoefficientsThe experimental results showed that, in administration period, the rat’s cardiac coefficients of each group showed no significant difference (P>0.05). Compared with the administration period, each group showed a great reduction(P<0.05).6 Pulmonary Histomorphologycompared with the control group, each administration dosage group no difference between the lung tissue.Dose group and high dose group of alveolar walls with mild edema, and a small amount of blood capillary hyperemia, some with a small amount of inflammatory exudation, can recover after the drug was stopped. Compared with the control group, the total difference, all within the normal physiological range.Conclusions1 Long-term using Shizaotang Decoction and Shizaotang Decoction mixed with licorice can cause cells in rats with acute cardiac injury, for the performance of AST, CK,CK-MB,LDH, a-HBDH increased.2 Shizaotang Decoction and Shizaotang Decoction mixed with licorice had slight side effects of lung tissue of rats. While they had a certain degree of injury to the cardiac muscle cells, the degree of damage is closely related to drug dosage.3 The side effect of euphorbia kansui, spurge, yuanhua, liquorice four herbs used together on rat-cardiopulmonary chronic toxicity depended on the dose factor. The factor of anti-classical prescription was not big, it just delayed the recovery of myocardial enzyme indicators.
