| Objective:To investigate the correlation between PARK16gene rs947211> rs823144polymorphisms and Parkinson’s disease (PD) and the differences in genotypic and allelic frequences between the Uygur and Han populations. Methods:Performed a case-control study, which included441PD patients (260cases of Han and181cases of Uygur) and495controls (287controls of Han and208controls of Uygur).Analyzed the genotypes and allele genotypes of PARK16gene rs947211> rs823144polymorphism sites by the methods of polymerase chain reaction (PCR-RFLP). Then verified the results by using the DNA sequencing method. Results:(1)rs947211A statistically significant difference was observed between the case group and the control of Han in genotypic and allelic frequencies (χ2=8.931, P=0.011and χ2=7.865, P=0.005), Individule who carried G allele had more risk than the non-carrier (OR=1.407,95%CI:1.108-1.788); There was also a statistically significant difference of genotypic and allelic frequencies in Uygur case group compared with the Uygur control group (χ2=8.100, P=0.017and χ2-7.117, P=0.008); The Uygur cases individule who carried G allele had more risk than the controls had (OR=1.471,95%CI:1.107-1.955).However, There was no statistically significant difference of genotypic and allelic frequencies in Han case group compared with the Uygur case group (χ2=0.053, P=0.974and χ2=0.013, P=0.911).(2)rs823144There were no statistically significant difference of genotypic and allelic frequencies in Han case group compared with the Han control group, Uygur case group compared with the Uygur control group, Han case group compared with the Uygur case group (χ2=1.519, P=0.468and χ2=1.592,P=0.207;χ2=0.325, P=0.850and χ2=0.173,P=0.678; χ2=2.173,P=0.337and χ2=2.315, P=0.128). Conclusion:There results suggest t that rs947211polymorphism of PARK16gene is associated with PD in the Uygur and Han populations. The G allele might be an independent risk factor and genetic marker for PD.However, there is no significant difference between Uygur with PD and Han with PD.There was no correlation between rs823144polymorphisms and PD. |
