Objective:To explore the association between the single-nucleotide polymorphism(SNP) ofthe rs3822086site of the α-Synuclein(SNCA)gene and Parkinson’s disease(PD)in XinjiangUygurs and Hans, to compare the distribution difference of this polymorphic site between theUygurs and Hans.Methods: The polymorphism of the rs3822086site of SNCA gene was determined bypolymerase chain-reaction restriction fragment length polymorphism(PCR-RFLP) in239patients with Idiopathic Parkinson’s disease (including93Uygurs and146Hans) and247health controls (including103Uygurs and144Hans) in Xinjiang region.Results:(1) In the age≥60years old group, the T/T genotype, C/T genotype and T allelefrequency of PD group (199cases) is higher than those in the control group(196cases)(26.1%vs20.9%ã€51.3%vs44.4%and51.8%vs43.1%), the polymorphismdistribution of SNCA gene have statistically difference between the two groups (genotype:χ~2=7.151,P=0.028, allele: χ~2=5.921,P=0.015,OR=1.416,95%CI=1.070~1.874). Furthercompared the T allele carriers(T/T+C/T) and C/C genotype frequency, the frequency of T/T+C/T genotype of PD group is obviously higher than that of the control group (77.4%vs65.3%), the difference is statistically significant (χ~2=7.056, P=0.008, OR=1.818,95%CI=1.167~2.833), Many factors binary logistic regression calibratied the confoundingfactors,such as nationality, gender, and age, there difference still have statistically significant(P=0.044, OR=1.515,95%CI=1.012~2.27).(2)Between the uygur and han nationalitypopulation,the T/T genotype, C/T genotype and T allele frequency of the uygurs is morelower than those of the hans(15.8%vs30.7%ã€44.9%vs50.3%and38.3%vs55.9%), thepolymorphism distribution of SNCA gene have statistically significance between the twopopulations (genotype: χ~2=28.979,P=0.000,allele: χ~2=28.989,P=0.000). Conclusion:(1) TheSNP of rs3822086site of SNCA gene meight be a potential susceptibility site of IPD patientswith the age≥60years in Xinjiang, rs3822086C/T+T/T is susceptible genotypes andrs3822086T is susceptible alleles.(2) the polymorphism distribution of rs3822086site ofSNCA gene meight be have significantly different between the Xinjiang Uygur and Hanpopulations. |
