| Objectives:The paper aims to investigate the expression of Hedgehog signaling pathway Gli-1and Smo factor in cervical cancer Siha cells following hypoxia and to study the impact of Hedgehog signaling pathway related factors on the biological behavior of cervical cancer Siha cells following hypoxia.Methods:The proliferation and migration of cervical cancer Siha cells following hypoxia was detected by MTT assay and wound healing assay. Following hypoxia cervical cancer Siha cells,the Gli-1-. Smo mRNA and protein expression were detected by RT-PCR and western blot.Results:In MTT assay and wound healing assay, Under different action time with hypoxia cervical cancer Siha cells, the cell proliferation and wound healing showed significantly raised than the control group. RT-PCR and Western-blot revealed that following hypoxia24hã€48h, the expression of HIF-1a gene and protein was significantly higher than the control group (P<0.05). The expression of Gli-1gene and protein was significantly higher than the control group (P<0.05). However, the expression of Smo gene and was no significant difference.Conclusion:Hypoxia will effectively enhance the cell proliferation and migration of cervical cancer Siha cell. Following hypoxia of cervical cancer Siha cells, gene and protein expression of HIF-1a and Hedgehog signaling pathway Gli-1obviously rose up. |
