Objective:To explore the effects of Esomeprazole(ESO)on the proliferation,migration,invasion and apoptosis of human cervical cancer SiHa cells and HeLa cells,and further explore its potential mechanism of action,so as to provide theoretical reference for the application of Esomeprazole in the treatment of cervical cancer.Methods:Normal cervical cell H8,Cervical cancer SiHa cells and HeLa cells were cultured in vitro.(1)CCK-8 assay was used to test the inhibitory effects of different concentrations of esomeprazole on the proliferation of cervical cancer SiHa cells and HeLa cells and Normal cervical cell H8 at 24,48 and 72h.(2)The effect of esmeazazole on the migration capacity of cervical cancer siha cells and hela cells was tested by cell picting.(3)The effect of esmeazazole on the ability of siha cells and hela cells was detected by transwell.(4)The apoptosis of the two cells of the cervical cancer siha cells and hela cells was treated by the flow cytometry.(5)Western blot was used to detect changes in the expression level of apoptosis related proteins.(6)Through western blot,the changes of the expression level of the pi3k/akt signaling pathway were further detected.Results:(1)CCK-8 assay showed that the proliferation of cervical cancer SiHa cells and HeLa cells was significantly inhibited after treatment with different concentrations(20,40,60,80,100μg/mL)of Esomeprazole(24h,48h,72h)compared with the control group(P<0.05).The proliferation of normal cervical H8 cells was not significantly affected(P>0.05).(2)Cell scratch test results showed that after different concentrations of esomerazole(60、80、100μg/mL)the mobility of SiHa cells and HeLa cells was significantly decreased with the increase of concentration and treatment time of esomeprazole(P<0.05).(3)Transwell experiment showed that with the increase of esomeprazole concentration,the number of SiHa cells and HeLa cells passing through the Transwell chamber decreased significantly(P<0.05).(4)Flow cytometry results showed that apoptosis rates of SiHa cells and HeLa cells in the experimental group increased significantly with the increase of drug concentration,with statistical significance(P<0.05).(5)Western Blot results showed that the expression levels of apoptosis-related protein Casepase3 were up-regulated,while the expression levels of Bcl-2 was down-regulated,with statistical significance(P<0.05).(6)Western Blot results showed that the expression levels of signaling pathway related molecules P-PI3K and P-Akt were significantly down-regulated(P<0.05),while the expression levels of PI3K and AKT were not significantly changed(P>0.05).Conclusion:1、Esomeprazole can effectively inhibit the proliferation,migration and invasion of cervical cancer SiHa cells and HeLa cells,and the inhibition gradually increased with the increase of concentration in a certain range.2、Esomeprazole promoted apoptosis of cervical cancer SiHa cells and HeLa cells by up-regulating casepasE3 and down-regulating Bcl2 expression levels.3、The inhibition of esomeprazole on cervical cancer SiHa cells and HeLa cells may be related to the inhibition of PI3K/AKT signaling pathway.
