Objective:To explore the FOXC2C-521-1and G-350-1, APOE and eNOS locus single nucleotide polymorphisms (SNPs) in yunnan Naxi correlation with type2diabetesMethods:Thesis select the naxi people in yunnan province, a total of513cases, including215cases of patients with type2diabetes,and the non-diabetes control298examples.By the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technology analysis FOXC2C-512T and G-350T, APOE gene polymorphism, using PCR technique to detect4introns eNOS gene polymorphism.Results:(1) in type2diabetes mellitus group:FOXC2C-512T the frequency of genotype CC, CT, and TT are18.1%,54%and18.1%;FOXC2G-350T the frequency of genotype GG, GT, and TT are68.4%,27.9%and68.4%;APOE gene E3/4, E2/2, E3/3, and E4/4the frequency distribution of14.4%,4.7%,80.9%and0%respectively;ENOS gene genotype4a,4b,4a/4b frequency are6%,79.1%and6%;All the distribution of genotype conforms to Hardy Weinberg equilibrium (P>0.05).(2) in type2diabetes FOXC2C-512T and G-350T, ApoE and eNOS gene polymorphism and clinical relevance:C-512T:type2diabetes mellitus group genotype frequency distribution between different ethnic groups found no statistical difference (P>0.05).G-350T:in diabetes group TC, HDL-C in GG and TT groups was statistically difference (P<0.05), TT is higher than the GG group.APOA, HBAIC in GG and GT group was statistically difference (P<0.05), GG group is higher than the GT.APOE:UA in E3/3and E2/2groups have statistical difference (P <0.05), E3/3genome than E2/2.ENOS27bp VNTR:type2diabetes mellitus group genotype frequency distribution between different ethnic groups found no statistical difference (P>0.05).(3) logistic regression analysis showed that FOXC2C-512T and G-350T, APOE and eNOS27bp VNTR gene polymorphism did not found a clear correlation in yunnan Naxi with type2diabetes (P>0.05).Regression analysis: waist circumference and LDL-C, uric acid is associated with type2diabetes (P<0.05).Conclusion (1) FOXC2C-512T and G-350T, APOE, eNOS gene polymorphism in yunnan naxi ethnic differences in the crowd.(2) FOXC2C-512T and G-350T, APOE, eNOS gene polymorphisms in metabolic index correlation;1) FOXC2C-512T:metabolic index was not found associated with C-512T gene polymorphism.(2) the G-350T:cholesterol, high-density lipoprotein cholesterol have correlation with G-350T gene polymorphism.(3) APOE;Uric acid has correlation with APOE gene polymorphism.(4) eNOS:metabolic index was not found associated with eNOS gene polymorphism.(3) FOXC2C-512T and G-350T, APOE and eNOS gene polymorphism in yunnan naxi population with type2diabetes mellitus has no obvious correlation between. |
