[Objective] To construct genetic basic data of the Naxi population living in Yunnan Province and providing a data basis for the application of the Y chromosome in forensic medicine, building terrace for personal identification and paternity cases of Naxi population.[Methods] Follow the principleofin-formedconsent, the collection stricted accordance with the criteria (unrelated male individuals, Naxi people within three generations, no blood lineage between individuals), we collected 5ml venous blood sample of 284 male participants from the Naxi settlement with EDTA-Na2 to avoid cruor in Lijiang, Yunnan. Genomic DNA was extracted using the Magnetic beads method of blood samples. The 35 Y-SNP loci were anaylsis by Mass ARRAY technology. The Naxi population data was performed using SPSS 17.0、Arleuin3.5、MEGA4.0、DISPAN、 JMP12.2.[Results] To build 2 multiple PCR panels analysing 35 Y-SNP markers. In the 284 unrelated male individuals, there only observed the mutation on the main trunkO、C、 D、F、K、N、P, the main trunk O included most of Naxi, accounting for about 45.4% of the sample. Based on the frequency of haploidO、C、D、F、K、N、Pand builded the phylogenetic tree, the principal component two-dimensional analysis diagram and clustering analysis diagram which displayed O haploid had high rate of mutation and relative, Language family features of clustering analysis in accordance with the population.[Conclusions] This study obtained allelic frequency distribution and statistical genetic parameters of forensic relevance of 35 Y-SNP loci in 284 unrelated male individuals. Constructed the Y chromosome database of Yunnan Naxi population, providing a data basis for the application in forensic medicine. To analysis Naxi population genetics, the genetic characteristics of ethic group as well as explanation of the observed genetic differences among them were comparatively understood, and holp to provid evidence for human evolution and biology of all ethnic origins, migration and dividing in China.
