Objective:To investigate the effect of Acitretin to Regulated upon activation normal T cell expressed and secreted protein (RANTES) and Monocyte chemoattractant Protein-1(MCP-1) in the peripheral blood and PBMCs of patients with psoriasis vulgaris, and Scored by PASI.To approach treatment mechanism of Acitretin to psoriasis, and the correlation between MCP-1and RANTES.Method:Sera and PBMCs were obtained from the venous blood sample from48patients with psoriasis vulgaris (before and after treatment of Acitretin) and30normal human controls.Enzyme-linked imunosorbent assay (ELISA) was used to examine the level of RANTES and MCP-1protein in the sera, quantitative real-time RT-PCR to detect the mRNA expression of RANTES and MCP-1in PBMCs. Psoriasis vulgaris Scored by PASI when before and after treatment of Acitretin.Results:PASI Score patency descend after the therapy of Acitretin in48Patients vulgaris (P<0.01). General recovery rate of Acitretin to psoriasis vulgaris was83.33%.The serum protein level and the mRNA expression in PBMCs of RANTES and MCP-1were significantly higher in Patients with Psoriasis vulgaris than those in normal group (P<0.01),and decreased after the treatment of Acitretin (P<0.01)The content of RANTES and MCP-1in serum have positive correlation with PASI Score when before and after treatment of Acitretin in ordinary psoriasis patients(r1=0.469, P<0.01; r2=0.431, P<0.01), The content of RANTES in PBMCs have positive correlation with PASI Score (r=0.141, P>0.05).But there is no apparently associativity between expression of MCP-1in PBMC and the difference of PASI Score.(r=0.484, P<0.01) Conclusion:Acitretin shows a patency clinical effect for psoriasis vulgaris, The treatment mechanism of Acitretin may have be positive correlation with RANTES and MCP-1of the CC chemotactic factor in peripheral blood. The content of RANTES in Peripheral blood have positive correlation with PAS I Score in ordinary psoriasis patients. The high expression of RANTES may be positive correlation with pathogenesis of psoriasis, RANTES may become one of index to evaluate pathogenetic condition of psoriasis vulgaris.
